白假丝酵母菌吡咯类药物相关耐药基因的突变和表达 被引量:4

摘要 目的了解白假丝酵母菌吡咯类药物相关耐药基因的突变和表达情况。方法采用裂解法提取白假丝酵母菌DNA,扩增ERG11基因后测序,选取3种吡咯类药物(氟康唑、酮康唑、咪康唑)均耐药的全耐药菌株DNA片段进行TA克隆;利用荧光定量PCR技术对CDR1、CDR2、MDR1基因表达的m RNA进行相对定量,比较耐药株与敏感株表达水平的差异。结果 37株耐吡咯类白假丝酵母菌中,ERG11存在41个碱基突变位点,其中20个同义突变,21个错义突变,发现新变异:N12I、Y13F、S16F、V36F、V51L、G129E、T123I、E194K、K344N、E517G、V234A;CDR1基因2-ΔΔCt值敏感株为1.09±0.27,耐药株为1.46±0.24,两者比较差异有统计学意义(P<0.05);敏感株与耐药株间CDR2和MDR1基因的表达水平比较差异无统计学意义(P>0.05)。结论白假丝酵母菌耐药机制较为复杂,可能与ERG11某些位点突变导致的氨基酸变异和CDR1高表达有关,CDR2和MDR1表达与耐药关系有待进一步研究。
