苍附导痰汤对肥胖型多囊卵巢综合征大鼠的瘦素、脂联素水平及胰岛素敏感指数的影响 被引量:18

摘要 目的:探讨苍附导痰汤对肥胖型多囊卵巢综合征大鼠的瘦素(Leptin,LPN)、脂联素(Adiponeetin,APN)水平及胰岛素敏感指数(Insulin Sensitivity Index,ISI)的影响。方法:采用改良Poresky造模法,制作肥胖型多囊卵巢综合征大鼠模型,造模后将大鼠随机分为模型空白组、二甲双胍组、苍附导痰汤组,每组15只,另选15只大鼠作为正常对照组。观察药物干预21d后各组大鼠体重、胰岛素敏感指数ISI、LPN、APN的变化,并计算。结果:与模型空白组比较,苍附导痰汤组和二甲双胍组大鼠体重、ISI、LPN水平均下降(P<0.01);APN水平上升;苍附导痰汤组APN水平变化较二甲双胍组更为明显(P<0.05)。与PCOS模型组相比,苍附导痰汤组大鼠血清T、LH水平均显著降低,E2显著升高,LH/FSH比值亦明显降低。与二甲双胍组比较,苍附导痰汤组降低LH水平效果更明显(P<0.01)。结论:苍附导痰汤可能通过降低体重、增加胰岛素敏感性、调节瘦素、脂联素和生殖激素的水平,从而改善大鼠生殖功能。 Objective: To explore effects of the Cangfu Daotan decoction on leptin, adiponectin level and insulin sensitivity index in obese rats with PCOS. Methods: Poresky method and high fat diet were used to build obese PCOS rat models. The PCOS rat models were randomly divided into model blank group, metformin group and the Cangfu Daotan decoction group, with 15 rats in each group. Furthermore, other 15 rats were used as normal control group. The changes of body weight, sex hormone, IRI, LPN and APN levels was observed and calculated in each group after the rats were given medicine for 21 days. Results: Compared with model control group, the weight,leptin,IRI levels decreased, and the APN levels increased in metformin group and the Cangfu Daotan decoction group (P〈0.01). The change of APN was more obvious in the Cangfu Daotan decoction group than that in metformin group (P〈0.05). The Cangfu Daotan decoction showed higher serum T, LH/FSH and lower E2 level compared with PCOS model group. LH level decrease in the Cangfu Daotan decoction group more obviously than that in metformin group (P〈0.01). Conclusion: The Cangfu Daotan decoction could improve the conception of rats by decreasing the weight, increasing insulin sensitivity and regulating the levels of leptin, adiponectin and sex hormone.
