摘要
目的探讨虎杖中白藜芦醇对人肝癌Hep G-2细胞株增殖和生长周期的影响。方法采用微波辅助法提取虎杖中白藜芦醇,将12.5,25.0,50.0,100.0μmol·L-1白藜芦醇作用人肝癌Hep G-2细胞,采用噻唑蓝(MTT)法检测虎杖中白藜芦醇对肝癌细胞Hep G-2细胞抑制增殖作用,在倒置相差显微镜下观察细胞形态变化,流式细胞术(FCM)检测细胞周期的分布。结果虎杖中白藜芦醇能显著抑制Hep G-2细胞的生长,并呈现时间-浓度依赖性。流式细胞仪结果显示,白藜芦醇作用于细胞Hep G-2且细胞被阻滞于S期,呈现出剂量依赖性。不同药物浓度白藜芦醇(12.5,25.0,50.0,100.0μmol·L-1)作用人肝癌Hep G-2细胞株48 h后,随着药物浓度的增大,G0/G1期的比例逐渐减小,与对照组比较差异有统计学意义(P<0.05)。而实验组细胞S期的比例随着浓度增大而增加,分别达到(44.82±1.21)%,(54.00±1.71)%,(65.21±3.66)%,(77.50±4.21)%,与对照组(34.75±3.92)%比较,差异有统计学意义(P<0.05)。结论虎杖中白藜芦醇能明显抑制肝癌Hep G-2细胞的增殖,并诱导其细胞凋亡,可引起肝癌Hep G-2细胞的S期阻滞。
Objective To investigate effects of resveratrol on the growth and cell cycle of human hepatoblastoma- derived HepG-2 cell line. Methods Resveratrol was extracted by microwave-assisted method and HepG-2 ceils were exposed to resveratrol at different concentrations ( 12.5,25.0, 50.0, 100.0 μmol·L-1 ). The viability of HepG-2 cells was measured by MTT. Morphologic changes of the cells were observed under the phase contrast microcope. The cell cycle of HepG-2 cells treated with resveratrol was analyzed by flow cytometry. Results Resveratrol could significantly inhibit proliferation of HepG-2 cells and increase the cells in the S phase in a dose-dependent manner. After 48 h treatment with different concentrations of resveratrol (12.5, 25.0, 50.0 and 100.0 μmol· L-1) , the cell proportion of G0/GI phase decreased as compared with control group (P〈 0.05). The cell proportion of S phase increased in a dose-dependent manner, ranging from (44.82 ± 1.21 ) % , (54.00 ± 1.71 )%, (65.21±3.66)% and (77.50±4.21)%, respectively, in the treatment groups as compared with negative control group[ (34.75e3.92)% ] (P〈0.05). Conclusion Resveratrol can inhibit the proliferation of HcpG-2 cells in vitro by inducing cell blockage at S phase.
