附子注射液对去卵巢骨质疏松大鼠骨代谢及骨密度的影响 被引量:5

Effects of Fuzi Injection on Bone Metabolism and Bone Mineral Density in Ovariectomized Rats with Osteoporosis

摘要 目的观察附子注射液对大鼠去卵巢骨质疏松模型骨代谢及骨密度(BMD)的影响。方法将60只清洁级Wistar雌性大鼠随机分为假手术组、模型对照组、附子小剂量组、附子中剂量组、附子大剂量组、阳性对照组,每组10只。模型对照组、附子小剂量组、中剂量组、大剂量组、阳性对照组切除卵巢。附子小、中、大剂量组分别给予附子注射液5,10,20 g·kg-1、阳性对照组给予雌二醇注射液0.05 g·kg-1,尾静脉注射,假手术组、模型对照组则分别给予同体积0.9%氯化钠溶液。均1次·d-1,连续8周。检测血清钙(Ca)、磷(P)、BMD、子宫质量指数、骨钙素(BGP)、碱性磷酸酶(ALP)、肌酐(Cr)、脱氧吡啶啉(DPD)、骨小梁厚度、骨小梁间距。结果与假手术组比较,模型对照组血清Ca、P水平、骨小梁间距、尿Ca/Cr、P/Cr、DPD/Cr水平均升高,差异有统计学意义(P<0.05或P<0.01),而BMD、子宫质量指数、BGP、ALP、骨小梁厚度均显著降低(P<0.01)。与模型对照组比较,附子大剂量组、阳性对照组血清Ca、P水平、骨小梁间距、尿Ca/Cr、P/Cr、DPD/Cr水平均降低(P<0.05或P<0.01)。而BMD、骨小梁厚度、BGP、ALP显著升高(P<0.01),且阳性对照组子宫质量指数显著升高(P<0.01)。结论附子注射液和雌二醇均能改善骨质疏松部分骨代谢生化指标,而大剂量附子注射液对子宫的刺激作用较雌二醇弱,其机制有待探究。 Objective To observe effects of fuzi injection on bone metabolism and bone mineral density (BMD) in ovariectomized rats with osteoporosis. Methods A total of 60 clean grade female Wistar rats were randomly divided into sham operation group ,model control group fuzi injection low-dose ,middle-dose ,high-dose group and positive control group with 10 rats in each group. Osteoporosis rat model was established in model control group,fuz/ injecton groups and positive control group. Fuzi injection groups receivedfuzi injection (5,10 and 20 g ~ kg-1 ) and positive control group received positive control (0.05 g ~ kg J ) by intravenous injection once daily for 8 weeks. Sham operation group and model control group were intravenously injected with the same volume of normal saline once a day for 8 weeks. Levels of serum calcium and phosphorus, BMD, uterus index, Bone gla protein ( BGP), alkaline phosphatase ( ALP ), creatinine ( Cr), deoxypyridinoline ( DPD ), rabecular thickness and trabecular space were examined. Results Compared with the sham operation group,levels of serum calcium,phosphorus,urine levels of Ca/Cr,P/Cr, DPD/Cr and trabecular space in model control group significantly increased (P〈0.05 or P〈0.01 ) ,while BMD, uterus index, BGP, ALP and trabecular thickness decreased significantly (P〈0.01). Compared with the model control group,levels of serum calcium, phosphorus,urine levels of Ca/Cr,P/Cr,DPD/Cr and trabecular space in bothfuzi group and positive control group decreased (P〈 0.05 or P〈0. 01 ), while BMD, BGP, ALP and trabecular thickness increased significantly ( P〈0. 01 ). Uterus index in positive control group increased significantly compared to the model control group (P〈0.01). Conclusion Both fuz/ and positive control injections can improve some biochemical indices of bone metabolism in osteoporosis. The uteral stimulation effect by high dose fuzi injection is lower than positive control and the mechanism needs further investigation.
