山楂总黄酮对高脂血症大鼠血管内皮功能的影响 被引量:4

The Effects of Haw Thorn Leaf Flavonids on Endothelial Function in Rats with Hyperlipidemia

摘要 目的:观察山楂总黄酮对高脂血症大鼠血管内皮功能的影响,并探讨其在降低血脂和保护血管内皮中的作用。方法30只大鼠随机分为对照组、高脂血症组和治疗组各10只。通过高脂、高胆固醇饮食建立高脂血症大鼠模型,对照组大鼠予以普通食料喂养;8周后治疗组大鼠继续高脂饮食的同时予以山楂黄酮(60 mg·kg-1·d-1)灌胃8周。16周末测定各组大鼠血脂、一氧化氮(NO)、内皮素-1(ET-1)、6-酮-前列腺素F1α(6-Keto-PGF1α)及丙二醛(MDA)含量的变化。结果与高脂血症组比较,16周末治疗组大鼠血浆甘油三脂(TG)、总胆固醇(TC)和低密度脂蛋白胆固醇(LDL-C)含量显著降低(P<0.05),而血浆高密度脂蛋白胆固醇(HDL-C)含量显著升高(P<0.05);16周末治疗组血浆ET-1、脂质过氧化产物MDA含量较高脂血症组明显降低(P<0.05),而血浆6-Keto-PGF1α、NO含量显著升高(P<0.05)。结论山楂总黄酮能有效地改善高脂血症大鼠脂代谢紊乱,同时具有降低脂质过氧化损伤,保护血管内皮细胞的功能。 Objective To observe the effect of haw thorn leaf flavonoids (HLF) on endothelial function in hyperlipidemic rats and to investigate the impact of HLF on decreasing cholesterol and endothelial protection. Methods 30 rats were divided into 3 groups randomly:10 rats in control group, 10 rats in hyperlipidemia group and 10 rats in treatment group. Hyperlipidemic rat models were produced by high-fat and high-cholesterol diet for 8 weeks. And control group were fed with normal diet. 8 weeks later,the treatment group were fed with high hyperlipidemic diet and HLF (60 mg·kg-1·d-1). At the end of 16 weeks, serum levels of lipid, Nitric oxide (NO), Endothelin (ET-1), 6-Keto-PGF1α and Malonaldehyde (MDA) were serially assessed. Results Compared with hyperlipidemia group at the end of 16 weeks, serum levels of triglyceride (TG), total cholesterol (TC) and LDL-C in treatment group decreased significantly (P〈0.05), but HDL-C increased (P〈0.05); and 16 weeks later ET-1 and lipid peroxidation (MDA) decreased (P〈0.05), but serum 6-Keto-PGF1α and NO production increased significantly (P〈0.05) in treatment group. Conclusion HLF has beneficial effects on lipid metabolism in hyperlipidemia rats, decreases the damage of lipid peroxidation, and improve endothelial function in hyperlipidemic rats.
