针康法对脑缺血大鼠运动功能及GAP-43、Nogo-A表达的影响 被引量:20

摘要 目的:探讨针康法对脑缺血大鼠运动功能及缺血区皮质GAP-43、Nogo-A表达的影响。方法:将SD大鼠随机分为5组,即针康组、针刺组、康复组、模型组和假手术组,按3天、7天、14天再分为3个亚组,每组6只。制备永久性脑缺血动物模型。针康组进行针康法干预;针刺组进行头穴丛刺治疗;康复组进行康复训练,假手术组与模型组不做任何干预。采用网屏试验评价各实验组大鼠的运动功能,免疫组化检测各实验组大鼠缺血区皮层GAP-43、Nogo-A的表达。结果:术后7天和14天,与康复组、针刺组比较,针康组网屏实验评分降低,差异有统计学意义(P<0.05);术后7天和14天,针康组缺血区皮层GAP-43阳性细胞表达高于模型组、康复组、针刺组(P<0.05);术后14天,针康组缺血区皮层Nogo-A阳性细胞表达降低,与模型组、康复组、针刺组相比差异有统计学意义(P<0.05)。结论:针康法能促进脑缺血大鼠运动功能的恢复,其机制可能与其促进脑缺血区皮层GAP-43及降低Nogo-A的表达有关。 Objective: To explore the effects of acupuncture - rehabilitation method on behavior and the expres- sion of GAP - 43 and Nogo - A in the cerebral cortex in rats after focal cerebral ischemia. Methods: A total of 90 SD rats were randomly divided into a model group, a sham operation group, an acupuncture with rehabilitation group, a rehabilitation group and an acupuncture group. Each group was subdivided into 3 subgroups by 3 d, 7 d, 14 d. The acupuncture- rehabilitation method was used in the acupuncture with rehabilitation group; tread- mill running was trained in the rehabilitation group; only the cluster needling of scalp point was punctured in the acupuncture group; the sham operation group and the model group had no intervention. Behavioral test of rats was evaluated by net screen test, and immunohistochemistry staining was used to observe the expression of GAP - 43 and Nogo - A in ischemic cortex in rats in each group at different time points. Results: After 7 d and 14 d, compared with the rehabilitation group and acupuncture group, the screen test score in acupuncture -re- habilitation group was decreased obviously, and the difference was statistically significant ( P 〈 0.05 ) ; after 7 d and 14 d, the expression of GAP -43 positive cells in ischemic cortex was higher in acupuncture - rehabilita-tion group than that in the model group, rehabilitation group and acupuncture group (P 〈 0.05 ) ; after 14 d, the expression of Nogo - A positive ceils in acupuncture - rehabilitation group in ischemic cortex was lower than that in the model group, rehabilitation group and acupuncture group, and the difference was statistically signifi- cant ( P 〈 0.05 ). Conclusion: The acupuncture - rehabilitation method can promote recovery of behavior in rats after focal cerebral ischemia and the mechanism may be related to increasing the expression of GAP -43 in cor- tex and decreasing the expression of Nogo - A in cortex.
