微生物末端限制性片段的长度多态性研究新进展

摘要 有效的比对不同环境下微生物群落基因组间的差异,可为刑事侦查中鉴定不同物证的来源或异同,提供有效的分析思路;微生物基因组16S rRNA有多个区段高度保守,可以设计出通用引物,扩增所有细菌的16S rRNA片段,而16S rRNA可变区的差异则可以用来区分不同的细菌;近年来发展起来的T-RFLP(末端标记限制性片段长度多态性)技术为该策略的实施提供了新的技术平台;本文对近年来T-RFLP分析的研究进展进行了系统回顾,对其关键技术,包括引物及限制酶的选择、末端片段的解析、信号的区分以及图谱的解读等方面的最新研究方法进行了系统的比较和评价,以期为其在法庭科学中的实践提供相应的参考和帮助。
