摘要
目的:研究不同浓度结缔组织生长因子(CTGF)及其抗体对鸡肌腱滑膜细胞增殖活性的影响。方法:以来亨鸡屈趾肌腱腱周滑膜组织为材料,经体外分离、培养和鉴定后,加入不同浓度的CTGF(浓度为0,5,10,20,50,100,200,500ng/ml)及CTGF抗体(浓度为0,1,2,4,8,16,32,64μg/ml)分组培养滑膜细胞,并以CCK-8染色法检测滑膜细胞的增殖情况。结果:滑膜细胞增殖在CTGF浓度为5ng/ml至200ng/ml时,呈逐步递增趋势,各组之间差异有统计学意义(P<0.05);而在200ng/ml及500ng/ml浓度组间差异无统计学意义(P>0.05),加入浓度为4,8,16,32μg/mlCTGF抗体时,滑膜细胞增殖呈逐步递减趋势,各组之间差异有统计学意义(P<0.05);而CTGF抗体浓度为0,1,2μg/ml和浓度为32,64μg/ml时,各组间差异无统计学意义(P>0.05)。结论:CTGF可促进体外培养的鸡肌腱滑膜细胞增殖,最佳浓度为200ng/ml;CTGF抗体对滑膜细胞增殖活性可产生一定程度的抑制作用。
Objective:To investigate the effect of connective tissue growth factor (CTGF) and its an- tibody on proliferation activity of tendon synovial cells. Methods:The tendon synovial cells were ob- tained from leghorn tendon synovial tissues and cultured with different concentration of CTGF and its antibody. Cell Counting Kit-8 was used to detect proliferation activity of synovial cells. Results: The proliferation activity of synovial cells increased gradually as the concentration of CTGF eleva- ting from 5 ng/ml to 200 ng/ml, significant tration (P〈0.05). However, no significant ly di higher than that in the control group at each concen fference was observed between synovial cells cultured with 500 ng/ml CTGF and those cultured with 200 ng/ml CTGF. And the proliferation acti synovial cells decreased gradually as the concentration of CTGF antibody elevating from 4 μg vtty /ml of to32 μg/ml, significant and those activity of significantly lower than that in the control group at each concentration (P〈0. 05). no difference was observed between synovial cells cultured with 64 μg/ml CTGF antibody cultured with 32μg/ml CTGF antibody. Conclusion. CTGF can promote the proliferation synovial cells in vitro with a optimal concentration of 200 ng/ml. And CTGF antibodycan inhibit the proliferation activity of synovial cells .
