紫外分光光度法测定氨茶碱中毒方法的建立与应用

摘要 目的建立血清中氨茶碱定量分析的紫外分光光度法,探讨对氨茶碱中毒患者实际应用的价值,为临床氨茶碱中毒提供诊疗依据。方法取0.5 ml血清加0.2 ml的0.1 mol/L盐酸溶液,再加入三氯甲烷-异丙醇(95∶5,V/V)的萃取液4.0 ml。充分振荡萃取,高速离心,取有机层于另一试管中加0.1 mol/L氢氧化钠溶液2.0 ml混匀反提,高速离心后取氢氧化钠层50μl进行紫外扫描。结果血清中氨茶碱的最大吸收峰为275 nm±1 nm,浓度在5.0μg/ml^60μg/ml呈线性关系,回归方程为y=0.0101x+0.0065,其相关系数为0.9998,方法的回收率为85.5%~102.4%,相对标准偏差(RSD)为3.5%~4.8%。日内、日间相对标准偏差(RSD)分别为3.08%~5.00%和3.16%~4.95%,最低检出浓度为2.0μg/ml。结论该方法操作简便、分析快速,结果准确,为临床诊断氨茶碱中毒提供了一个简便准确的检测方法。 Objective To establish an UV spectrophotometry method for the quantitative analysis of aminophylline in serum,and to explore the application value for aminophylline poisoning patients,so as to provide evidence for clinical treatment and diagnosis of aminophylline poisoning. Methods 0. 1 mol / L hydrochloric acid solution of 0. 5 ml in serum and 0. 2 ml,add chloroform- isopropyl alcohol( 95∶5,V / V) extract 4. 0 ml. After Shake Extraction,high speed centrifugation,removed the organic layer into another test tube with 0. 1 mol / L sodium hydroxide solution to 2. 0 ml for blending anti stripping,after high speed centrifugation,extracted sodium hydroxide layer 50 μl for UV scanning. Results The absorption peak of serum aminophylline was( 275 ± 1) nm,concentration is linear between 5. 0 μg / ml and 60 μg / ml,y = 0. 0101 x + 0. 0065,the correlation coefficient is 0. 9998,recovery rate was 85. 5% ~ 102. 4%,the relative standard deviation( RSD) was 3. 5% ~ 4. 8%. The relative standard deviation( RSD) were 3. 08% ~ 5. 00% and 3. 16% ~ 4. 95% respectively,the minimum detectable concentration was2 μg / ml. Conclusion This method has advantages of simple operation,fast detection and accurate results.,which provides a simple and accurate method for clinical diagnosis of aminophylline poisoning.
