黄连素联合谷氨酰胺对小鼠癌性恶病质模型的干预 被引量:2

摘要 目的探讨黄连素联合谷氨酰胺对癌性恶病质模型的改善作用及机制。方法使用小白鼠结肠腺癌细胞株CT-26皮下注射BALB/c小白鼠,构建荷瘤小白鼠肿瘤恶病质模型,将小白鼠分为5组:A组(无荷瘤小白鼠组)、B组(荷瘤无干预组)、C组(荷瘤黄连素干预组)、D组(荷瘤谷氨酰胺干预组)和E组(荷瘤黄连素联合谷氨酰胺干预组)。C组小白鼠给予黄连素灌胃[25 mg/(kg·d)],D组小白鼠给予谷氨酰胺灌胃[25 mg/(kg·d)],E组给予黄连素[25 mg/(kg·d)]联合谷氨酰胺[25 mg/(kg·d)]灌胃。连续2周记录小白鼠饮食量、体质量,测量并比较各组肿瘤瘤体质量、生化指标、血清白细胞介素1(interleukin 1,IL-1)、白细胞介素6(interleukin 6,IL-6)、肿瘤坏死因子(tumor necrosis factor,TNF)差异,观察黄连素联合谷氨酰胺对癌性恶病质疾病进展的干预效果。结果 C组、D组和E组相对于B组可显著改善荷瘤小白鼠的体质量减轻程度,改善生化指标,并降低血清中细胞因子IL-1、IL-6及TNF表达水平,差异有统计学意义(P<0.05)。其中E组指标更优于C、D组。结论口服黄连素联合谷氨酰胺可以有效改善癌性恶病质小鼠状态,其机制可能与降低细胞因子表达水平有关,需进一步研究。 Objective To investigate the improvement effect and the mechanism of berberine in conjunction with glutamine in treating cancer cachexia. Methods Tumor-bearing mouse models with cancer cachexia were constructed u- sing BAL]3/c mice by subcutaneous injection of mouse C-26 colon cancer cell lines.These mice were divided into 5 groups: Groups A (non-tumor-bearing mice), B(tumor-bearing mice without intervention), C (tumor-bearing mice with berberine intervention),D (tumor-bearing mice with glutamine intervention) and E (tumor-bearing mice with intervention of ber- berine in conj unction with glutamine).Berberine, glutamine, and berberine in conj unction with glutamine, at the same dos- age [25 mg/(kg./d)] were given by gavage in mice in groups C,D and E respectively.Their food intakes and body masses were recorded for two consecutive weeks.Differences in tumor masses,biochemical indices,serum interleukin I (IL-1) ,IL- 6 and tumor neucrosis factor (TNF) were measured and compared among all groups.Intervention effects of berberine in conjunction with glutamine on progression of cancer cachexia were observed. Results Compared with group B,decrease in body mass and biochemical indices were significantly improved and expression levels of serum eytokines(IE-1, IL-6 and TNF)were decreased in tumor-bearing mice in groups C,D and E (P〈0.0B).Indices in group E were superior to those in groups C and D. Conclusion Oral administration of berberine in conjunction with glutamine can improve conditions of cancer-caehectic mice effectively.The possible mechanism is related to decreased expression of cytokines,whieh should be further studied and extended.
