Relation between GST-π,Helicobacter Pylori and Gastric Carcinoma

摘要 目的研究不同胃黏膜病变组织中谷胱甘肽巯基转移酶π(glutathione S-transferase-π,GST-π)蛋白表达情况,探讨GST-π蛋白与幽门螺杆菌(helicobacter pylori,Hp)及胃癌(gastric carcinoma,GC)的相关性。方法选取慢性萎缩性胃窦炎、胃窦溃疡及胃窦癌各40例,采用免疫组织化学链霉菌抗生物素蛋白-过氧化物酶连结(streptavidinperosidase,SP)法与免疫蛋白印记法(Western blot)对GST-π蛋白在不同胃黏膜病变中表达进行检测;吉姆萨(Giemsa)染色检测GC组织Hp感染情况。结果 1GST-π蛋白在GC组织中表达阳性率明显高于其它组(85%,P<0.05);2与其它组织相对比,GST-π蛋白在GC组织中呈现高表达(P<0.05);3与阴性组织相对比,GC阳性组织GST-π蛋白表达更高(P<0.05)。结论 GST-π蛋白与GC的发生具有相关性,Hp感染可能促进GST-π表达更多,两者对于临床上GC的诊断都具有重要意义。 Objective To investigate the relation between GST-Л,helicobacter pylori and gastric carcinoma by stud ying the expression of GST-Л in different gastric mueosal lesions tissues.Methods The Immunohistochemistry Technique (SP method) and Western blot were utilized to detect the expressions of GST -Л in different gastric mucosal lesions tissues (including atrophic gastritis,gastric ulcer and gastric carcinoma in 40 cases).Infection of helicobacter pylori was tested by Giemsa staining experiments.Results The positive rate of GST-Л protein expression was significantly higher than the other groups in gastric carcinoma tissue (85% P d0.05).Compaed with other tissues, the expression of GST-~r protein was up-regulation in gastric carcinoma (57.17 ± 6.51, P 〈0.05).Compared with the tissue of negative group, the ex- pression of GST-Л protein was higher in the tissue of helicobacter pylori infecting group (58.88 ± 6.71, P〈0.05).Conclu- sion The GST-Л protein is related to the occurrence of gastric carcinoma, and the infection of helicobacter pylori may promote the expression of GST-Л, which are related to the clinical diagnosis of gastric carcinoma significantly.
