摘要
GmFT2a是大豆光周期反应中的关键基因,为研究其表达调控的分子机制,以光周期敏感品种自贡冬豆和钝感品种黑河27为材料,比较分析两品种Gm FT2a启动子序列的差异,发现在自贡冬豆GmFT2a启动子区存在两个含有光反应元件的插入片段T1和T2,而黑河27无上述片段。根据光响应元件I-box和Sp1的序列,构建诱饵载体,利用酵母单杂交技术筛选自贡冬豆叶片cDNA文库,获得一个与T1元件结合的MYB转录因子。该转录因子的获得为进一步研究GmFT2a表达调控的分子机制提供了新的线索。
Gm FT2 a is a key gene in photoperiodic pathway of soybean. However,the underlying molecular mechanism of Gm FT2 a expression is not clear. In this study,a photoperiod- sensitive soybean cultivar Zigongdongdou and insensitive cultivar Heihe 27 were used as materials to compare the Gm FT2 a promoter sequences. It revealed that there were 2 insertion / deletion( In Del) fragments,T1 and T2,which contained light reaction elements in the Gm FT2 a promoter of Zigongdongdou. However,the same fragments were not in that of Heihe 27. The bait constructs were prepared according to the sequences of light reaction elements of I- box and SP1 to screen c DNA library of Heihe 27 leaves,and a MYB transcription factor bound with T1 was isolated with the yeast one- hybrid system. The obtaining of the MYB transcription factor provided a favorable clue for further study of the molecular mechanism Gm FT2 a expression regulation.
