黑地黄丸对5/6肾切除大鼠肾衰竭模型血液动力学的影响 被引量:5

摘要 目的:通过观察黑地黄丸对5/6肾切除大鼠肾衰竭模型血液动力学的影响,探讨该方对肾脏的保护机制。方法:70只Wistar大鼠随机分为7组:空白组,模型组,尿毒清组、洛丁新组,黑地黄丸高、中、低剂量组。除空白组外,其他各组大鼠行5/6肾切除术,建立肾衰竭大鼠模型,分别连续给药12周后测定血浆血管内皮素(ET)、降钙素基因相关肽(CGRP)、血管紧张素I(ATⅠ)、血管紧张素II(ATⅡ)。结果:与尿毒清组比较,黑地黄丸高剂量组大鼠血浆ATⅡ含量降低(P<0.05),ET、ATⅠ含量显著降低(P<0.01);与模型组比较,黑地黄丸高组剂量组大鼠血浆CGRP含量显著升高(P<0.01)。结论:高剂量黑地黄丸能降低肾衰竭大鼠血浆ET含量,升高血浆CGRP水平,减少ATⅠ、ATⅡ合成,有效改善肾衰竭大鼠的血液动力学,对肾衰竭大鼠的肾功能具有保护作用。 Objective:To explore the protective mechanism of Heidihuang pill on the kidney by observing the hemodynamic influences of the medicine on renal failure rat model with 5/6 nephrectomy. Methods:70 Wistar rats were randomly divided into 7 groups:blank group,model group,Niaoduqing group,Lotensin group and Heidihuang pill of high,medium and low dose group. Except the blank group,the rats of other groups were performed 5/6 nephrectomy to establish renal failure rats model and were given medicine respectively for 12 consecutive weeks. After 12 weeks,the content of plasma vascular endothelin(ET),calcitonin gene related peptide (CGRP), angiotensin Ⅰ(AT Ⅰ) and angiotensin Ⅱ(AT Ⅱ) were measured. Results:Com-pared with Niaoduqing group, the AT II contents of the Heidihuang pill of high dose group decreased (P<0.05) and the ET and AT I content decreased significantly(P<0.01);compared with model group,the CGRP level of Heidihuang pill of high dose group increased significantly (P<0.01). Conclusion:high dose of Heidihuang pill can reduce renal failure rats’ plasma ET content,increase plasma CGRP content and re-duce AT Ⅰ,AT Ⅱ synthesis,which shows effectively hemodynamic improvement for the renal failure rats and protection for kidney functions of the renal failure rats.
