摘要 比较了不同培养基(碳源、氮源、无机盐)以及不同发酵条件(温度、pH、摇床转速、接种量、装液量)对海洋红酵母003菌株发酵产量的影响。通过正交试验确定了003菌株的最佳的培养基:葡萄糖1.0%,玉米粉0.1%,蛋白胨0.4%,酵母膏0.5%,NaCl 1.0%,自然pH。海洋红酵母003菌株最佳摇瓶发酵条件为:温度22℃,摇床转速200r/min,装液量250mL瓶装50mL,接种量8%,发酵时间24h。在此条件下,菌量可达1.02×109 cfu/mL。对海洋红酵母003菌株进一步进行5L发酵罐扩大培养试验,在接种量为8%、初始pH 4.8、搅拌速率280~350r/min、通气量6L/min条件下,菌量可达1.05×109 cfu/mL。 Effect of medium ingredient including carbon source,nitrogen source,inorganic salt and culture condition of temperature,pH,shaking speed,inoculation volume,fluid volume on the yield of marine red yeast 003 were investigated.The results by orthogonal test showed that the maximal biomass was obtained at glucose 1.0%,corn flour 0.1%,peptone 0.4%,yeast extract 0.5% and NaCl 1.0% at the natural pH.Submerged culture was carried out in a 250 mL flask containing 50 mL of medium with 8% inoculum and incubated at 200r/min for 24 hand 22 ℃.Under the above condition,the amount of bacteria could reach 1.02×109 cfu/mL.The cell biomass of was obtained at1.05×109 cfu/mL when the marine red yeast was cultured in a 5Lfermentor with 8%inoculum at pH4.8and 22 ℃ stirred at 280-350r/min and the air ventilation volume were 6L/min.
