大蒜素对膀胱癌细胞增殖和B细胞淋巴瘤-2表达的影响 被引量:3

摘要 目的探讨大蒜素对膀胱癌BIU-87细胞的增殖及B细胞淋巴瘤-2(Bcl-2)表达的影响。方法用不同浓度的大蒜素处理膀胱癌BIU-87细胞36h,以噻唑蓝(MTT)比色法分析细胞生长抑制率,逆转录聚合酶链式反应(RT-PCR)检测Bcl-2 mRNA的表达,蛋白免疫印迹技术检测Bcl-2蛋白表达的变化。结果与对照组比较,经大蒜素处理的BIU-87细胞的生长抑制率增加(P<0.05),且效应呈浓度和时间依赖性(P<0.05);大蒜素处理细胞后,BIU-87细胞中Bcl-2 mRNA及蛋白质的表达水平下调(P<0.01)。结论大蒜素能抑制BIU-87细胞的增殖,且呈剂量和浓度依赖性,其机制可能与大蒜素下调Bcl-2mRNA和蛋白的表达有关。 Objective To explore the influence of allicin on proliferation of BIU-87 cells of bladder cancer and expression of B-cell lymphoma-2( Bcl-2). Methods Different concentrations of allicin were used to process BIU-87 cells for 36 hours. Growth inhibitory rate of cell was evaluated by MTT assay,the mRNA expression of Bcl-2 was detected by RT-PCR,and protein expression of Bcl-2 was detected by Western blot. Results Compared with control group,the growth inhibition rate increased in BIU-87 cells processed by allicin( P〈0. 05),and the effect was dependent on concentration and time. The mRNA and protein expressions of Bcl-2 decreased significantly in BIU-87 cells processed by allicin( P〈0. 01). Conclusion Allicin can inhibit the growth of BIU-87 cells,the effect is dependent on concentration and time,and its mechanism may be correlated with down-regulation of Bcl-2 mRNA expression and protein by allicin.
