黄淮麦区部分小麦品种(系)高分子量麦谷蛋白亚基的SDS-PAGE和分子标记分析 被引量:12

摘要 利用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)和分子标记技术,对黄淮麦区47份小麦育种材料高分子量麦谷蛋白亚基(HMW-GS)进行鉴定和分析。SDS-PAGE结果表明,在所检测的小麦品种(系)中,Glu-A1位点编码的HMW-GS有3种类型,分别是Null、1和2*,其中1出现频率较高(80.9%),Null次之(17.0%),2*仅有1份;Glu-B1位点有7+8、7+9、17+18共3种类型,其中7+8和7+9出现频率较高,分别为48.9%和44.7%;Glu-D1位点有2+12、5+10、4+12共3种类型,其中5+10出现频率最高(61.7%)。利用Dx5、Ax2*、By8、By9和By17亚基特异性分子标记检测结果表明,参试材料中含各标记亚基的材料依次为29(61.7%)、1(2.1%)、23(48.9%)、21(44.7%)、3(6.4%)份;在所检测的小麦品种(系)中含最优亚基组合Dx5、By8的材料共13份,频率为27.7%。分子标记检测结果与SDS-PAGE检测结果相吻合,表明亚基特异性分子标记可用来快速检测小麦材料中的HMW-GS基因。 The types of high molecular weight glutenin subunits (HMW-GS) of 47 wheat materials in Huang Huai wheat area were identified and analyzed by sodium dodecyl sulfate polyacrylamide gel e- lectrophoresis (SDS-PAGE) and molecular marker technology. The results of SDS-PAGE showed that three alleles-encoded HMW-GS (Null, 1 and 2* ) were identified at Glu-A1 locus with the highest fre- quency (80. 9%),followed by Null (17.0%),2* was lowest (only 1 cultivar);3 alleles at GIu-B1 (7+8,7+4-9,17+18)with the higher frequency (48.9%) at subunit 7--8(44.7%) at subunit 7+ 9 ; 4 alleles atGlu-D1 (2+12,5@10,4+12)with the highest frequency (61.7%) at subunit 5--10. Dx5 , Ax2 * ,By8 ,By9 and By17 genes of tested wheat cuhivars were detected by specific molecular mark ers,and the number of wheat varieties (lines) contained corresponding genes were 29 (61. 7%), 1 (2.1%) ,23(44.7%),21(44.7%) and 3(6.4%),respectively. 13 varieties (lines) with optimal sub unit combinations (Dx5 and By8 ) were identified with frequency of 27. 7% in all tested varieties (lines). The detection results of molecular markers and SDS-PAGE detection showed that the subunit specific molecular markers could be used for rapid detection of HMW-GS gene in wheat materials.
