不同剂量生山药煎液对脾虚便秘模型小鼠的胃肠道影响

Effect of Shanyao Decoction on Spleen Deficiency Constipation Model Mice Defecation and Intestinal Vasoactive Intestinal Peptide VIP and Substance P

目的:通过研究生山药煎液对脾虚便秘小鼠排便功能、小肠推进率的改变及血管活性肠肽(VIP)、P物质(SP)含量的影响,观察不同剂量的生山药煎液治疗脾虚便秘的疗效,进而阐明其治疗便秘的机理,并对量效关系作初步探讨。方法:将60只清洁级SPF小鼠随机分为空白组,模型组,生山药煎液高、中、低剂量组,麻仁丸组。经造模成功后,除模型组外,各组小鼠分别用凉开水、山药水煎高、中、低剂量及麻仁丸进行灌胃处理。末次给药后,观察山药煎液对小鼠排便功能、小肠推进率的影响,采用免疫组织化学法对SP﹑VIP进行定性及定量检测。结果:与模型组比较,不同剂量的山药煎液有促进小鼠排便的作用,以山药水煎剂高剂量组明显(P<0.01);各给药组均可促进便秘小鼠小肠内墨汁的推进速度,以山药煎液高剂量组作用最为显著(P<0.01),其次为山药煎液中剂量组、麻仁丸和山药煎液低剂量组(P<0.05);山药煎液高、中、低剂量组和麻仁丸组对脾虚便秘模型小鼠肠组织中SP﹑VIP含量的改变均有显著性差异(P<0.05)。结论:山药水煎高、中、低剂量组和麻仁丸组小鼠的排便功能、精神状态、大便质地均有明显改善,其小肠内墨汁有显著的推进,SP、VIP含量均较模型组降低。

Objective： By studying the impact of Shanyao Decoction on intestinal propulsion and vasoactive intestinal peptide（ VIP） P substance（ SP） content in spleen constipation mice to observe the efficacy of different doses of Shanyao Decoction and thereby to clarify the mechanism of the treatment of constipation and study the dose- effect relationship.Methods： 60 male SPF mice were randomly divided into control group,model group,Maren pill group,Shanyao Decoction with low,middle and high dose groups. After modeling success,except the model group,the mice were given Shanyao Decoction in high,middle and low doses and Maren Pills and water control treatment. We observed the effect on defecation function and small intestine and the indexes of SP and VIP were detected by immunohistochemical method. Results： The experimental observation showed that the high,middle and low doses of Shanyao Decoction and Maren Pills groups＇ defecation function,mental state and the stool texture were improved compared with those of the model group. All groups can improve the ink promoting speed in the small intestine and the Shanyao Decoction with high dose group was the most obvious（ P〈0. 01）. The contents of SP and VIP in high,middle and low doses of Shanyao Decoction and Maren Pills groups were significant. Conclusion： The high,middle and low doses of Shanyao Decoction and Maren Pills groups can improve the defecation,mental status and stool texture of mice with constipation and they can promote the speed of ink promoting in small intestine significantly and SP and VIP in intestinal tissue of mice were significantly decreased compared with those of model group.
