基于C57/BL6J小鼠的动物移植性肺癌模型的建立及评价研究 被引量:2

摘要 目的:建立皮下荷瘤移植的C57/BL6J小鼠肺癌模型。方法:采用培养细胞移植法,将已培养传代好的Lewis肺癌细胞株接种于C57/BL6J品系小鼠腋腹部皮下,3周后无菌条件下取出肿瘤,用套管针将肿瘤小块平均注射至小鼠腋腹部皮下,约为8mm3/只。2周后进行实验研究,测量小鼠体质量、血细胞、淋巴细胞和脾重。结果:与无瘤对照组比较,荷瘤组的体质量增长较快,白细胞数升高,红细胞数减少,相应的血红蛋白的含量降低,CD3、CD4、CD8细胞含量均有上升,CD4/CD8比值降低,NK细胞(CD49阳性)含量下降,荷瘤组的平均脾重是无瘤组的4倍多。结论:利用Lewis肺癌细胞株可以建立皮下种瘤移植的荷瘤小鼠肺癌模型,为移植性肺癌模型的建立和应用提供实验依据。 Objective: To establish the C57 / BL6 J mice lung cancer model of subcutaneous tumor transplantation. Methods: By using cell- culture transplant method,cultured and passed down well Lewis lung cancer cells were inoculated into mice axillary subcutaneous abdominal C57 / BL6 J strains. 3 weeks later,the tumor was taken out after under aseptic- condition removed,trocar tumor small average- injection to mice axillary subcutaneous abdominal,about 8 mm3 for each.Experimental study was performed 2 weeks later. The weights,blood cells,lymphocytes and the spleen weight of the mice were measured. Results: Compared with no tumor group,tumor group's weight growth,elevated white blood cell count and red blood cell number decreased. The content of the corresponding hemoglobin was reduced. CD3,CD4 and CD8cells were increased and CD4/ CD8 decreased. NK cells( CD49 positive) content decreased and the average weight of spleen tumor group was 4 times more than that of the non tumor group. Conclusion: The use of Lewis lung cancer cell lines can be used to establish lung carcinoma bearing mice model of subcutaneous tumor transplantation and provide the experimental basis for the establishment and application of transplanted lung cancer model.
