亚麻子提取物对四氯化碳致小鼠急性肝损伤的保护作用 被引量:4

摘要 目的:探讨不同浓度亚麻子水提物与醇提物对四氯化碳(CCl4)致小鼠急性肝损伤的保护作用。方法:90只小鼠随机分成9组:空白对照组、CCl4模型组、联苯双酯组及亚麻子水提物和醇提物低、中、高剂量组,各组均连续灌胃给药7 d后灌胃0.35%四氯化碳(CCl4),染毒24 h制备小鼠急性肝损伤模型,计算肝指数,测定血清谷丙转氨酶(ALT)和谷草转氨酶(AST)活性、肝组织中丙二醛(MDA)含量以及过氧化物歧化酶(SOD)活性,并观察肝组织HE染色切片的病理变化。结果:亚麻子提取物低、中、高剂量组均能显著降低CCl4致急性肝损伤小鼠ALT、AST活性,升高肝匀浆中SOD活力,降低MDA含量,与CCl4模型组比较,差异有高度统计学意义(P<0.01)。病理切片表明与模型组比较,不同浓度亚麻子提取物组均能使小鼠肝损伤减轻。结论:亚麻子水提物与醇提物对CCl4所致小鼠急性化学性肝损伤有保护作用,其机制可能与抗脂质过氧化有关。 Objective: To study the protective effect of water extract and ethanol extract of linseed on carbon tetrachloride( CCl4)- induced acute hepatic injury in mice. Methods: A total of 90 mice were randomly divided into 9 groups:control group,CCl4 model group,bifendate group,low,medium and high dose of water extract of linseed groups( equivalent drug 1,3,5 g / kg),low,medium and high dose of ethanol extract of linseed groups( equivalent drug 1,3,5 g / kg). All groups were lavaged for 7 d. The model of acute hepatic injury in mice could be set up by gavage 0. 35% CCl40. 1 m /10 g,exposed for 24 h. The liver indexes were calculated. The activities of serum ALT and AST,hepatic SOD and level of hepatic MDA were also determined. At the same time,histopathological changes of liver tissue slices stained by HE staining were observed. Results: The activities of ALT,AST and level of hepatic MDA were significantly decreased by low,medium and high dose of extract of linseed while activity of SOD in liver tissue was markedly increased. Compared with the CCl4 model group,there were significant differences( P〈0. 01). The histopathological results also showed the obvious improvement of liver injury in different concentrations of water extract and ethanol extract of linseed groups. Conclusion: The water and ethanol extracts of linseed have protective effect on acute hepatic injury induced by CCl4 in mice.The mechanism may be related with the inhibition of lipid peroxidation.
