摘要
目的应用固相微萃取-气相色谱/质谱联用技术(SPME-GC/MS)检测分析体外培养肠杆菌的挥发性有机物(VOCs),探讨SPME-GC/MS检测应用于肠杆菌菌种鉴别的可行性。方法取肠杆菌40株,培养24 h后,通过SPME-GC/MS技术收集并检测分析细菌培养产生的VOCs。结果 40株肠杆菌检测出浓度较高的VOCs有14种,而乙酸在大肠埃希菌菌种的检出率为100%,在其余菌种中检出率为0,3-甲基-1-丁醇在肺炎克雷白菌种、十四烷醇酯在产气肠杆菌中检出率均为100%,在其余菌种中检出率均为0。3-甲基-1-丁醇,乙酸、吲哚,十四烷醇酯分别为肺炎克雷白菌、大肠埃希菌、产气肠杆菌的相对特异性VOCs。结论通过SPME-GC/MS技术可检测到部分肠杆菌的特异性VOCs,将其应用于肠杆菌菌种的鉴别具有可行性。
Objective To detect the volative organic compounds(VOCs) of enterobacteriaceae cultured in vitro with solid-phase micro-extraction and gas chromatography-mass spectrometry(SPME-GC/MS) technique.Methods Forty samples of enterobacteriaceae were collected and cultured in the mycobottles for 24 hours,and the VOCs of the bacteria were detected and analyzed by SPME-GC/MS.Results Fourteen main VOCs were detected from all bacteria,and the detection rate of acetic acid was 100%in escherichia cob' group and 0 in other enterobacteriaceae groups.The detection rate of 1-butanol 3-methyl was 100%in klebsiella pneumoniae group and the detection rate of 1,14-Tetradecanediol was 100%in enterobacter aerogenes group,but the detection rate was 0 in other enterobacteriaceae groups.1-Butanol,3-methyl might be the specific VOCs for klebsiella pneumoniae,Acetic acid and Indole might be the specific VOCs for escherichia coli,and 1,14-Tetradecanediol for enterobacter aerogenes.Conclusions The specific VOCs of enterobacteria can be detected with SPME-GC/MS technique.The application of SPME-GC/MS technique in identifying enterobacteriaceae is feasible.
