摘要
目的探讨家兔急性肺栓塞(APE)对血清生长分化因子-15(GDF-15)的影响及其意义。方法健康日本大耳白兔29只,随机分为APE组(n=16)及对照组(n=13),通过自体血栓输入复制APE模型,对照组注入生理盐水。采用多导生物信号采集系统监测右心室收缩压及舒张压(RVSP和RVDP)、颈动脉收缩压及舒张压(SBP和DBP),直至APE后8 h,于注血栓(水)前、后2、4、6及8 h抽血,通过酶联免疫法(ELISA)检测血清GDF-15及氨基末端脑钠肽前体(NT-pro BNP)的浓度,注水后及APE后2、4和8 h各处死3只动物行右心室心肌电镜检测。结果 APE后右心室心肌细胞出现线粒体肿胀、破坏、肌纤维断裂;GDF-15的浓度于APE 6 h后高于APE前(P<0.05),GDF-15浓度与RVSP、SBP及NT-pro BNP浓度呈线性相关,相关系数R为0.76(P<0.05)。结论 APE后GDF-15血清浓度明显升高,并与病情的严重程度相关。
[ Objective ] To explore the effect of acute pulmonary embolism(APE) in rabbits on growth differentiation factor 15(GDF-15). [ Methods ] Twenty-nine rabbits were divided randomly into two groups: embolism group(n =16) and control group (n =13). The rabbit model of APE was induced by injecting antologous embolus. The rabbits of control group were injected with saline instead of embolus. The right ventricle systolic pressure (RVSV), right ventricle diastolic pressure(RVDP), systolic blood pressure(SBP) and diastolic blood pressure (DBP) were surveyed by the multi-biological signal acquisition system. The serum concentrations of GDF-15 and pro-B-Type natriuretic peptide (NT-proBNP) were determined by enzyme linked immunosorbent assay before and 2 h, 4 h, 6 h and 8 h after injecting clots or saline. Every three rabbits in control group and 2 h, 4 h and 8 h after APE were killed to illustrate the pathological changes of right ventricle myocardium by transmission electron microscopy, respectively. [ Results ] Mitoehondria swelling, rupture and myocardial fiber disorder could be seen in right ventricle myocardium after APE. The serum concentration of GDF-15 significantly increased 6h after APE (P 〈0.05). It had linearity correlation to NT-proBNP, RVSP and SBP (R =0.764, P 〈0.05). [ Conclusion ] The increased serum concentration of GDF-15 is obvious after APE. The concentration of GDF-15 probably relates to severity of APE.
