摘要
右旋糖酐酶按2.5 U/300 m L与肠膜明串珠菌种子液同时加入发酵培养基,反应24 h后可定向合成重均分子量集中在10~100 ku之间的右旋糖酐。发酵液经0.45μm和100、50、30、10 ku分级膜滤可分别获得符合药用级别要求的重均相对分子质量为67、42、20 ku的右旋糖酐产品。与传统方法相比,酶法耦合分级膜法可有效控制右旋糖酐分子量分布,产品纯度高、产率高、反应条件温和,能耗低、成本低、易于实现工业化。
Dextranase 2.5 U/300 m L add to fermentation medium with seed liquid of Leuconosto mesenterondes. Main weight-average molecular weight between 10 ku to 100 ku of dextran be rational synthesized after 24 h. Fermentation broth is separated by multi-grade membrane with bore diameter 0.45 μm, 100, 50, 30 and 10 ku. The weight-average molecular weight of dextran are 67, 42 and 20 ku which are conform to medicinal dextran. Compared with traditional methods, this method which used Enzymatic couple multi-grade membrane can control the molecular weight distribution of dextran effectively. The purity and productivity of dextran are high. This method has advantages of mild reaction conditions, low energy consumption, low cost, and is suitable for industrial production.
