摘要
目的:研究五倍子酸(GA)对人纤维肉瘤HT1080细胞增殖和凋亡的影响。方法:分别用3.125、6.250、12.500、25.000、50.000和100.000 mg/L的GA处理HT1080细胞48 h后,应用MTT法检测细胞的抑制率;分别以6.250、12.500和25.000 mg/L的GA作用HT1080细胞48 h后,应用流式细胞仪检测细胞凋亡情况,应用免疫细胞化学方法检测GA对HT1080细胞中Bcl-2和Bax蛋白表达的影响。结果:GA对HT1080细胞以剂量依赖的方式抑制其增殖(F=71.214,P<0.001)、诱导其凋亡(F=52.217,P<0.001),并能上调Bax蛋白的表达(F=11.024,P<0.001)、下调Bcl-2蛋白的表达(F=8.741,P<0.001)。结论:GA可能通过上调Bax蛋白的表达和下调Bcl-2蛋白的表达来抑制HT1080细胞增殖和诱导其凋亡。
Aim: To explore the inhibitory effects of gallic acid( GA) on human fibrocarcinoma cell HT1080.Methods: MTT was employed to detect proliferative ability in HT1080 cells treated with 3.125,6.250,12.500,25.000,50.000 and 100.000 mg / L GA for 48 h,respectively.Flow cytometry was used to examine apoptotic rate in HT1080 cells treated with 6.250,12.500 and 25.000 mg / L GA for 48 h,respectively.Immunocytochemistry was applied to explore the change of Bcl-2 and Bax expression in HT1080 cells.Results: The proliferation inhibition rate of HT1080 cells was enhanced in a dose-dependent manner( F = 71.214,P〈 0.001).The number of apoptosis cell increased in a dose-dependent manner( F = 52.217,P〈 0.001).GA up-regulated Bax expression( F = 11.024,P〈 0.001) and down-regulated Bcl-2expression( F = 8.741,P〈 0.001).Conclusion: GA probably inhibit the proliferation and induce apoptosis of HT1080 cells by up-regulating Bax and down-regulating Bcl-2.
