富血小板血浆可注射组织工程骨促进兔下颌牵张成骨实验研究 被引量:1

Promotion of bone regeneration by an injectable tissue engineered bone constructed with platelet-rich plasma in rabbit mandibular distraction osteogenesis

摘要 目的探讨富血小板血浆可注射组织工程骨对兔下颌骨牵张成骨新骨形成的促进作用。方法将40只新西兰白兔随机分为A组、B组、C组、D组各10只。建立下颌骨牵张成骨动物模型,固定期第2天,A组于牵张间隙注射富血小板血浆可注射组织工程骨,B组注射富血小板血浆和纤维蛋白胶原的混合液,C组注射纤维蛋白胶原,D组注射生理盐水作为空白对照组。分别于固定2、6周时处死动物获取标本,通过大体观察、HE染色、X线观察、骨密度分析观察骨质愈合改建情况。数据进行单因素方差分析和组间两两比较的SNK-q检验,P<0.05为差异有统计学意义。结果通过大体观察、HE染色、X线观察、骨密度分析显示,在固定期第2、6周,A组、B组牵张间隙内成骨质量好于C组、D组,A组优于B组。A组固定第2、6周牵张区新生骨骨密度值分别为(66.74±1.97)、(94.65±2.35)g/cm2,B组分别为(43.63±1.84)、(68.54±2.03)g/cm2,C组分别为(27.15±1.49)、(53.69±1.64)g/cm2,D组分别为(25.39±1.23)、(55.32±1.52)g/cm2,A组牵张区内新生骨骨密度明显高于B组、C组、D组(均P<0.05),B组明显高于C组和D组(均P<0.05)。结论富血小板血浆可注射组织工程骨可有效促进兔下颌骨牵张成骨过程中的新骨形成。 Objective To investigate the promoting effects of an injectable tissue engineered bone constructed with platelet-rich plasma on bone regeneration in rabbit mandibular distraction osteogenesis. Methods Forty New Zealand 's white rabbits were randomly divided into four groups,and 10 rabbits for each group. Distraction osteogenesis model was made in these rabbits. On the second day of the consolidation period,group A,B,C and D were respectively injected an injectable tissue engineered bone constructed with platelet-rich plasma,the mixture of platelet-rich plasma and fibrin glue,fibrin glue and physiological saline at distraction gap. At the 2nd and 6th week of consolidation,rabbits were sacrificed to evaluate bone regeneration by gross observation,hematoxylin and eosin staining,radiographic examination and bone mineral density analysis. ANOVA and SNK-q test were used to process the data,the result of P〈0. 05 was considered statistically significant. Results At the 2nd and 6th week of consolidation,the ossification quality of distraction gap was better in group A and B than in group C and D,and group A was superior to group B through gross observation,hematoxylin and eosin staining,radiographic examination and bone mineral density analysis. The bone mineral density in distraction area was( 66. 74 ± 1. 97) g/cm^2and( 94. 65 ± 2. 35) g / cm^2 respectively in group A at the 2nd and 6th week of consolidation; and( 43. 63 ± 1. 84) g/cm^2and( 68. 54 ± 2. 03) g / cm^2 respectively in group B; and( 27. 15 ± 1. 49) g / cm^2and( 53. 69 ± 1. 64)g / cm^2 respectively in group C; and( 25. 39 ± 1. 23) g / cm^2and( 55. 32 ± 1. 52) g / cm^2 respectively in group D. The bone mineral density in distraction area was significantly higher in group A than in group B,C and D( P〈0. 05),and the bone mineral density in distraction area was significantly higher in group B than in group C and D( P〈0. 05). Conclusions The injectable tissue engineered bone constructed with platelet-rich plasma can effectively promote bone regeneration in rabbit mandibular distraction osteogenesis.
