豆渣中可溶性膳食纤维提取的研究 被引量:10

摘要 以大豆豆渣为原料,先用传统化学方法碱法处理得到大豆可溶性膳食纤维(SDF)和不可溶性膳食纤维(IDF),然后再用改进的酶法处理前一步得到的不可溶性膳食纤维,进一步提取大豆可溶性膳食纤维,并通过单因素试验及正交试验对碱法和酶法条件进行了优化。湿豆渣经烘干、粉碎、碱液水解、酶解、沉淀、干燥后制得膳食纤维。结果表明,碱法制备可溶性膳食纤维的最佳工艺条件是:温度80℃,物料比1∶15,反应时间1.5h,p H13。在此条件下,豆渣中SDF得率为18.2%。碱处理得到的IDF使用复合多糖酶处理法提取可溶性膳食纤维的最佳工艺条件是:温度45℃,物料比1∶15(m∶v),加酶量10.0%,反应时间1.5h,p H4.5。在此条件下,SDF得率为11.09%。 The extraction technology of soybean soluble dietary fiber(SDF)and insoluble dietary fiber(IDF)from bean dregs were investigated by alkaline solution,then the extraction technology of soybean soluble dietary fiber(SDF)from insoluble dietary fiber(IDF)were also investigated by enzyme hydrolysis. The single factor experiment and orthogonal experiment were designed to obtain optimized technology. Dietary fiber was prepared from wet soybean dregs by the processing of drying,crushing,soaking with alkaline solution,enzymatic hydrolyzing,precipitating and drying. The results showed that the optimum alkaline solution conditions were 80 ℃,p H 13,alkaline solution 1.5h,ratio of substrate to water 1 ∶ 15. Under optimal conditions,the yield of soluble dietary fiber was increased to 18.2%. And the results of optimum enzymatic hydrolysis conditions were :45℃,p H 4.5,enzyme concentration 10.0%,reaction time1.5h,ratio of substrate to water 1 ∶ 15. Under the optimal conditions,the yield of soluble dietary fiber was increased to 11.09%.
