摘要
目的:建立测试银花感冒冲剂中绿原酸含量的高效液相方法,并对此方法进行系统的方法学验证,以确保应用该方法测试的结果准确、可靠。方法:采用色谱柱:Agilent TC-C18柱(4.6 mm ×150 mm,5μm,Agilent,美国),流动相:甲醇-0.5%磷酸溶液(15∶85),检测波长为327 nm。结果:绿原酸在25~800μg/mL范围内线性关系良好,得到线性回归方程为Y=27.88X+2.128,相关系数为0.9998(n=6);低、中、高浓度精密度的RSD值分别为0.59%、0.58%、0.44%;重复性RSD为0.26%;低、中、高浓度的加样回收率分别为99.24%、99.43%、99.75%;供试品溶液室温放置8 h稳定, RSD为0.29%。结论:本方法测定银花感冒冲剂中绿原酸的含量准确、可靠,操作简单、用时短,可用于银花感冒冲剂中绿原酸的含量测定。
Objective:To establish a high performance liquid phase method to measure the content of chlorogenic acid of Flos Lonicerae in Ganmao granules,and to ensure that the results are accurate and reliable by systematic methodology.Methods:The chromatographic column:Agilent TC-C18 column (4.6 ×150 mm,5 μm,Agilent,American),a mobile phase of methanol:-0. 5%phosphoric acid solution (15:85),the detection wavelength was 327 nm.Results:The chlorogenic acid in 25ug mL-1 -800ug· mL^-1 linear relationship was good,the linear regression equation was Y =27.88X+2.128,correlation coefficient was 0.999 8 (n =6);low,high concentrations of RSD precision,values were 0.59%,0.58%,0.44%;repetitive RSD 0.26%;low,recover-ies.The high concentration were respectively 99.24%,99.43%,99.75%;the test solution was placed at room temperature for 8 h,and RSD was 0.29%.Conclusion:This method for determination of the content of chlorogenic acid in Flos Lonicerae cold gran-ule is accurate and reliable with simple operation and short time.It can be used for the determination of chlorogenic acid in Flos Lonicerae in Ganmao granules.
