摘要
目的探讨芪药鸡金粥对化疗大鼠胃肠黏膜机械屏障损伤的保护作用,从ERK/MAPK通路的角度探讨机制。方法将48只SPF级Wistar大鼠随机分为药膳组(药膳灌胃)、模型组(白粥灌胃)、空白组(白粥灌胃)各16只,于实验第7天,药膳组和模型组行氟尿嘧啶150mg/kg一次性腹腔注射,制备胃肠黏膜损伤大鼠模型。化疗后第1天和第8天,每组各随机抽取8只大鼠处死后取回肠组织,采用蛋白质印迹法检测相关指标。结果化疗后第1天,模型组连接蛋白显著下降(均P<0.05),空白组各通路蛋白的表达水平显著或相对低于同期药膳组和模型组水平(均P<0.05);化疗后第8天,药膳组Claudin-1连接蛋白表达显著高于模型组(P<0.05),各通路蛋白表达显著高于同期空白组(均P<0.05),模型组ERK1/2蛋白表达显著高于空白组(P<0.05)。结论芪药鸡金粥可调节大鼠肠道机械屏障紧密连接蛋白的表达,从而保护机械屏障的结构与功能,可能是通过ERK/MAPK通路的活化而发挥保护作用。
Objective To explore the effect of Chinese herbal medicated congee on mediating the mechanical barrier function of intestinal epithelial cells in rats with chemotherapy-induced gastrointestinal mucositis,and to study the mechanism through the ERK/MAPK pathway.Methods Forty-eight SPF-grade Wistar rats were randomly assigned to one of three groups,with 16 rats in each group.The intervention group was fed with Chinese herbal medicated congee,while the model group and the control group were given rice porridge.On the 7th day of study,rats in the intervention group and the model group were injected intraperitoneally with 5-fluorouracil at 150mg/kg to produce gastrointestinal mucositis.Eight rats were selected from each group respectively on the 1st and 8th day after chemotherapy and were sacrificed to collect the intestinal tissues,then related parameters were measured by western blotting.Results The tight junction protein expression in the model group significantly dropped on the 1st day after chemotherapy,and the expression levels of all of the channel proteins in the control group were relatively or significantly lower than the other two groups(P0.05 for all).On the 8th day,Claudin-1expression in the intervention group was significantly higher than that in the model group(P0.05),and the values of all of the channel proteins in the intervention group were significantly higher than the control group(P0.05 for all).The results showed that the expression level of ERK1/2was significantly increased in the model group compared to the control group(P0.05).Conclusion Chinese herbal medicated congee adjusts the expression of tight junction proteins,thus to protect the structure and function of the intestinal barrier.The effect may be achieved by mediating the ERK/MAPK pathway.
