摘要 为研究DF-1细胞的保存期及其传代过程中的稳定性和安全性,将来源于ATCC的DF-1细胞建立细胞库,取液氮保存12、24及48个月的DF-1细胞复苏,经3次传代后,观察不同批次细胞的生长特性并接种鸡传染性法氏囊病病毒,研究各批次细胞出现病变的时间及病毒含量;同时制备不同代次细胞的染色体并进行核型分析;对基础细胞库14代和最高代次60代的DF-1细胞进行致瘤性检测。结果显示,DF-1细胞在液氮中保存48个月,对细胞生长特征及增殖病毒无明显影响,经传60代后的细胞,染色体核型无明显差异且无致瘤性,表明DF-1细胞在传代过程中是稳定且安全的。 In order to understand the stability and safety for DF-1 cell during passage and storage, cells derived from ATCC were recovered and cultured from cell bank which were conserved in nitrogen canister for 12,24 and 48 months respectively. Following three-time passages,growth characteristics for each batch of DF-1 ceils were recorded, and then inoculated with infectious bursal disease virus, time for cytopathogenesis appearing was recorded and virus titer was calculated. At the same time,chromosome samples were prepared and karyotype analysis was carried out for DF-1 cells with different passages. Tumorigenicity for the 14th generation which was the foundation cell bank and the maximum generation (the 60th generation) of ceils were test. The results showed that 48-month conservation in nitrogen canister had no significant impact on cell growth and virus replication, and DF-1 ceils passaged 60 times had no difference in karyotype and had no tumorigenicity. It was concluded that DF-1 cell was highly stable and safe during passaging.
