四物汤治疗更年期综合征的作用机制研究

Study on the mechanism of Siwutang decoction in the treatment of climateric syndrome

目的观察四物汤对更年期模型SD大鼠的作用并探讨发挥作用的可能靶点和机制。方法将正常SD雌性大鼠40只按体质量均衡和随机的原则分为正常组、模型组、四物汤低剂量组、四物汤中剂量组、四物汤高剂量组。除正常组外,其余组均切除两侧卵巢造模。手术2周后,正常组和模型组给予等量蒸馏水灌胃,四物汤低剂量组、四物汤中剂量组、四物汤高剂量组分别给予3,6,9 g/(kg·d)四物汤灌胃,均1次/d,持续6 d。第7天腹主动脉取血并分离血清,分离子宫并称质量,免疫组化法测定子宫组织雌激素受体ERα表达量;蛋白质免疫印记技术检测药物血清对T47D细胞雌激素受体ERα表达的影响。结果正常组大鼠的子宫系数与模型组比较差异有统计学意义(P<0.01),四物汤低剂量组、四物汤中剂量组、四物汤高剂量组的子宫系数与模型组比较差异均有统计学意义,且随着四物汤方剂浓度的升高,子宫系数有增大的趋势;大鼠子宫组织ERα表达模型组与正常组比较差异有统计学意义(P<0.01),四物汤中剂量组和高剂量组与模型组比较差异有统计学意义(P<0.01);四物汤低、中剂量组的含药血清能够明显提高T47D细胞ERα的表达水平(P均<0.01)。结论四物汤对更年期综合征有很好的治疗作用,并可提高靶细胞ERα的表达水平。

Objective It is to observe the effect of Siwutang decoction on climateric syndrome in SD rat models and to ex- plore the possible target point and mechanism. Methods Forty female SD rats were randomly divided into five groups： normal group, model group and low, medium and high concentration groups. The rats in all the groups except normal group were re- sected on both sides of the ovary to establish models. In 2 weeks after operation, normal group and model group was given the same volume of distilled water by intragastric administration, and low, medium and high concentration groups were treated with Siwutang decoction at the dosage of 3,6, 9 g/（ kg·d） respectively once per day and for 6 days. On the seventh day, animals were sacrificed and uteri were separated solely and weighed. The blood was collected, and serum was separated. The expres- sions of subtypes-estrogen receptor-alpha （ERQ） on the uteri were detected by immunohistoehemical method. The effect of the pharmacological serum on proliferation assay with human breast cancer cell line （T47D） was detected by immune protein im- printing technology. Results There were significant differences in the uterus coefficient between normal group and model group （ P 〈 0.01 ） . The uterus coefficient of three Siwutang groups were different compared with model group, and with the increase of concentration of Siwutang decoction prescription, uterus coefficient increased accordingly. In the expressions of ERa on the uteri, there were significant differences between normal group and model group（ P 〈 0.01 ） and there were also differences a- mong model group and medium concentration group and high concentration group. Low concentration of Siwutang decoction could increase the expressions of ERa in the T47D cells（ P 〈 0.01 ）. Conclusion Siwutang has a good effect on menopausal syndrome, and can improve the expression of ERα of target cells.
