川芎嗪对急性心肌缺血模型大鼠离体心脏心功能的影响 被引量:4

摘要 目的观察川芎嗪对急性心肌缺血模型大鼠离体心脏心功能的影响并探讨其作用机制。方法将40只急性心肌缺血造模成功大鼠随机分为对照组、普奈洛尔组、异丙肾上腺素组、川芎嗪+普奈洛尔组及川芎嗪组,于造模成功24 h后断头取心,用Langendorff离体心脏灌流方法,对照组灌流KH营养液,其余各组灌流相应药物,测定5组大鼠在药前10min及药后0.5 h、1 h心肌收缩力(IT)、心率(HR)、左室内压力峰值(LVSP)及左室射血分数(LVEF)、左室最大上升/下降速率(±LVdp/dtmax)。结果在灌流给药0.5 h、1 h,川芎嗪组IT、LVSP、LVEF、HR及±LV dp/dtmax明显增高(P均<0.05),与对照组及给药前比较,差异均有统计学意义(P均<0.05)。结论川芎嗪可增强急性心肌缺血模型大鼠离体心脏心肌收缩力,加速传导,改善缺血心脏心功能,对急性心肌缺血再灌注损伤心肌具有一定的保护作用。 Objective It is to observe the effect of Ligustrazine on cardiac function of isolated heart of rat models with a- cute myocardial ischemia and to explore its mechanism. Methods 40 successful acute myocardial ischemia rat models were randomly divided into control group, propranolol group, isoproterenol group, ligustrazine + propranolol group and ligustrazine group. The rats heads were cut off and their heart were gotten in 24 h after model successful establish. The control group was given KH nutrient solution, and the other group was given respective drugs by Langendorff isolated heart perfusion method, I- sometric tension(IT) , heart rate (HR) , left ventricular peak pressure (LVSP) and left ventricular ejection fraction (LVEF) , left ventricular maximum increase/decrease rate (LVdp/dtmax) were detected in five groups at 10min before medication, at 0.5 h, 1 h after medication. Results At 0.5 h, 1 h after medication,IT, LVSP, LVEF, HR and dp/dtmax + LV significant- ly increased in Ligustrazine group , and compared with that in the control group and before administration, the differences were all significant ( P 〈 0.05 ). Conclusion Ligustrazine can enhance heart myocardial contractility of isolated heart of the model rats with acute myocardial ischemia, accelerate conduction, improve the function of ischemic heart, and has a protective effect on myocardial reperfusion injury after acute myocardial ischemia.
