Brachyury is a founding member of T-box transcription factor family. Recently, accumulating evidence demonstrated that Brachyury is closely related to tumor progression, and plays a key role in the process of epithelial-mesenchymal transition (EMT), suggesting Brachyury may be a potential target for T cell- mediated immunotherapy of cancer. This review summarized the relationship between Brachyury and tumor metastasis, staging, and prognosis, the induction and regulation of EMT mediated by Brachyury, and the role of Brachyury in the resistance to traditional therapy of cancer and cancer immunotherapy targeting Brachyury. It also gived fresh perspectives on further investigation and clinical application of Brachyury.
