宫颈鳞癌组织中血管生成拟态及HIF-1α的表达 被引量:3

摘要 目的研究宫颈鳞癌组织中是否存在血管生成拟态(VM),以及其与缺氧诱导因子-1α(HIF-1α)与宫颈鳞癌发生发展浸润转移的关系和临床意义。方法收集2010年1月至2011年12月泸州医学院附属医院经病理证实为人宫颈鳞癌组织石蜡标本67例,进行CD34/过碘酸雪夫染色(PAS)双重染色检测是否存在VM;采用免疫组化检测HIF-1α在宫颈鳞癌组织中的表达,分析VM与HIF-1α的相关性及临床病理特征。另随机选择同期因子宫肌瘤行子宫全切手术的正常宫颈组织标本20例作为对照。结果宫颈鳞癌组织中VM和HIF-1α阳性率分别为38.81%(26/67)、64.18%(43/67),显著高于正常宫颈组织[0(0/20)],差异均有统计学意义(P<0.05);不同病理分级、国际妇产科联盟(FIGO)分期及有无淋巴结转移患者宫颈鳞癌组织中VM阳性率比较,差异均有统计学意义(P<0.05)。不同肿瘤大小及有无淋巴结转移患者HIF-1α阳性率比较,差异也均有统计学意义(P<0.05)。VM与HIF-1α呈正相关(r=0.339,P<0.05)。结论宫颈鳞癌组织中存在VM、HIF-1α高表达可能促进VM的形成,可能共同促进宫颈鳞癌的浸润和转移。 Objective To study whether the vasculogenic mimicry in cervical squamous cell carcinoma tissues exists,as well as its relationship and clinic pathological parameters between hypoxia-inducible factor-1α(HIF-1α) protein and invasion and metastasis of progress of cervical squamous cell carcinoma. Methods A total of 67 cervical squamous cell carcinoma resection specimens confirmed by pathology from January 2010 to December 2011 in the Affiliated Hospital of Luzhou Medical College were collected and detected whether it existed VM by CD34/PAS dual immunohistochemical staining. It adopted immunohistochemical staining to detect the expression of HIF-1α protein in cervical squamous cell carcinoma tissues and analyze the association between VM and HIF-1α,as well as its clinic pathological characters. In addition,it selected 20 normal cervical tissues specimens underwent total hysterectomy for uterine fibroid as the control group. Results In the cervical squamous cell carcinoma tissues,the positive expression rate of VM and HIF-1α was respectively 38.81%(26/67),64.18%(43/67),which was distinctively higher than that in the normal cervical tissues[0(0/20)]. The difference had statistical significance(P〈0.05);The formation of VM was associated with the FIGO staging, histology grading and lymphatic metastasis. The difference had statistical significance(P〈0.05).The positive expression of HIF-1α protein was associated with lymphatic metastasis and cancer size,which was statistically significant in difference(P〈0.05). There was a positive association between the expression of HIF-1α and VM(r =0.339,P 0.05).Conclusion VM exists in cervical squamous cell carcinoma tissues. Overexpression of HIF-1α may induce the formation of VM.VM and jointly promote the invasion and metastasis of cervical squamous carcinoma.
