卡维地洛对大鼠颈动脉损伤后VSMCs迁移和增殖的影响

摘要 目的探讨卡维地洛(CAR)对大鼠颈动脉损伤后VSMCs迁移、增殖和内膜增生的影响。方法 Wistar大鼠54只,随机分为假手术组、损伤组、CAR组,每组18只,除假手术组外各组建立大鼠颈动脉球囊损伤模型。3组均于术后第7、14、28 d处死大鼠。观察颈动脉形态学变化,免疫组织化学检测增殖细胞核抗原(PCNA)的表达。结果假手术组,无新生内膜发生。与假手术组比较,损伤组7、14 d和28 d VSMCs大量迁移、增殖,内膜面积(IA)显著增加,有统计学意义(P<0.05),且随着损伤时间的延长而逐渐增加,而管腔面积(LA)显著减少(P<0.05)。与损伤组比较,CAR组从中膜迁移至内膜的VSMCs明显减少,IA均低于相同时间点损伤组,有统计学意义(P<0.05),且在损伤后14 d抑制作用最显著,而管腔面积显著扩大(P<0.05)。结论 CAR可有效抑制血管损伤后VSMCs迁移、增殖和内膜增生。 Objective To study the effect of carvedilol (CAR) on the VSMCs migration and proliferation and neointimal formation in rats with injured carotid artery. Methods 54 Male Wistar rats were randomly divid- ed into the sham - operated group, the injury group, and the CAR group( n = 18). The model established by bal- loon injury of rat carotid arteries except the sham - operated group. Carotid tissues of three groups were harvest- ed 7 days~ 14 days and 28 days after injury. The target vascular sections were stained with I-IE to study the mor- phology changes. The expression of PCNA in arterial wall was detected by immunohistochemistry. Results No neointima was found in the sham- operated group. Compared with the sham- operated group, on 7th,14th and 28th day after balloon injury, a large number of VSMCs migrated and proliferated in the injury group. The inti- mal area (IA) increased significantly(P 〈 0.05) and gradually increased with time prolong after injury, while lumen area (LA) had a significant reduction(P〈 0.05). Compared with the injury group, the intimal VSMCs number migrating from media reduced significantly, the IA were lower at the same time point and its value were the most significant at 14th day (P〈 0.05), while the lumen area at the same time point enlarged significantly (P 〈 0.05) in the CAR group. Conclusions Carvedilol may inhibit VSMCs migration, proliferation and neointimal formation in rats with injured carotid artery.
