摘要
目的以放射敏感性不同的鼻咽癌细胞株CNE-1和CNE-2为研究对象,分析经不同剂量X射线照射后鼻咽癌细胞株CNE-1和CNE-2蛋白质的拉曼光谱。比较不同剂量X射线对两种鼻咽癌细胞株蛋白质的影响。方法应用细胞集落实验分析CNE-1和CNE-2细胞的放射敏感性;激光共聚焦显微拉曼光谱仪检测分析射线照射后CNE-1和CNE-2细胞蛋白质的变化。结果 CNE-1细胞的放射抗性(存活分数SF2)是CNE-2细胞的1.43倍。拉曼光谱技术分析结果表明,两种细胞株蛋白质的拉曼光谱谱峰基本一致,在归属为L-苯丙氨酸的952cm-1位置,经照射CNE-1和CNE-2细胞株谱峰强度不同;两种细胞株的L-苯丙氨酸在照射后72h的拉曼谱峰强度显著增强,提示此时L-苯丙氨酸的含量有所增加。结论CNE-1和CNE-2细胞放射敏感性存在着差异;探寻放射敏感性的特征拉曼标志,将可作为研究肿瘤放射敏感性预测的手段之一。
Objective To investigate the influences of different doses of X-ray irradiation on CNE-1and CNE-2proteins of nasopharyngeal carcinoma(NPC)cell lines with different radiosensitivity,by Raman spectroscopy(RS).Methods Radiosensitivity of CNE-1and CNE-2was measured and compared by clonogenic survival assay.The changes of the proteins of CNE-1and CNE-2after irradiation were analyzed by RS.Results The radioresistance(in terms of survival fraction,SF2)of CNE-1cells was 1.43 times of that of CNE-2cells.The Raman spectra exhibited similar spectral profiles with subtle differences.The intensity of the peak at 982cm-1 assigned to L-phenylalanine was different for irradiated CNE-1and CNE-2and increased significantly 72 hafter X-ray irradiation.Conclusion The radiosensitivity of CNE-1and CNE-2is different.Raman characteristic target of cell radiosensitivity may provide a promising means for tumor radiosensitivity prediction.
