摘要
目的研究老年扬子鳄和幼年扬子鳄气管中BMP-4与PAR-2表达的年龄相关变化。探讨BMP-4、PAR-2与扬子鳄气管衰老之间的关系,为进一步研究爬行动物呼吸系统衰老机制积累材料。方法采用HE染色法和免疫组织化学SABC法分别对老、幼扬子鳄气管切片进行显色,光镜观察并进行统计分析处理。结果 BMP-4与PAR-2阳性细胞在老、幼扬子鳄气管中都有很多分布。在老鳄气管壁中BMP-4与PAR-2阳性细胞分布于黏膜层、黏膜下层及外膜层中最外层的纤维膜,同时外膜软骨层的软骨囊及部分软骨细胞也有BMP-4与PAR-2表达,软骨细胞间质显色很少。在幼鳄气管壁中黏膜层、黏膜下层及外膜层中最外层的纤维膜有BMP-4与PAR-2阳性细胞分布,而整个外膜软骨层都几乎无显色。结论 BMP-4和PAR-2在气管壁软骨层中表达可能与扬子鳄气管衰老、病变有关。
Objective: To investigate the age-related changes of BMP-4 and PAR-2expressed in the trachea of Alligator sinensis, and explore the relationship between BMP-4,PAR-2 and tracheal aging to aim at providing basic information of the study on the aging mechanisms of reptilian respiratory system. Methods: The immunohisto-chemical SABC method and HE staining were used to observe the distribution of cells in the trachea under a light microscope,and statistic analysis was also used. Results: There were many BMP-4- and PAR-2-like immunoreactive cells in the tracheae of old and young Alligator sinensis. About the trachea of old Alligator sinensis, BMP-4- and PAR-2-like immunoreactive cells existed in the mucosa, submucosa and fibrosa, the two proteins also occurred in cartilage capsules and parts of cartilage cell nuclei, with small quantity in the interstitial of cartilage cells. About the trachea of young Alligator sinensis, BMP-4 and PAR-2 were positively expressed in the mucosa,submucosa and fibrosa, and were negative expressed in the whole cartilage layer. Conclusion: BMP-4 and PAR-2 expressed in the cartilage layer may be related to the aging and lesion of the trachea of Alligator sinensis.
