吡喹酮联合γ-干扰素治疗对日本血吸虫性肝纤维化小鼠Smads蛋白表达的影响 被引量:3

摘要 目的研究Smads蛋白在日本血吸虫病小鼠肝纤维化形成后,经过吡喹酮杀虫和γ-干扰素(IFN-γ)抗纤维化治疗后其在小鼠肝组织中的表达。方法以日本血吸虫尾蚴感染BALB/c小鼠构建肝纤维化模型,在感染16周后,将所有存活的小鼠随机分成三组:空白对照组、吡喹酮治疗组、吡喹酮联合IFN-γ治疗组,并以10只未感染的BALB/c小鼠作为正常对照。治疗8周后,取小鼠肝组织,进行病理学检测,观察小鼠肝组织中胶原纤维沉积情况,判定肝纤维化程度;采用免疫组织化学方法,观察Smads蛋白在小鼠肝组织中的表达情况,并利用其阳性表达面积进行半定量分析。结果空白对照组小鼠肝纤维化程度计分为(6.90±1.57)分,显著高于正常对照组(1)分,经吡喹酮治疗后小鼠肝纤维化程度略有下降为(5.03±1.82)分,加用IFN-γ治疗后,小鼠肝纤维化程度较吡喹酮组有明显改善为(3.5±1.96)分(P<0.05),但未达到正常水平。免疫组化的研究提示,Smads蛋白主要表达在虫卵肉芽肿周围及肝窦区。吡喹酮治疗组以及吡喹酮联合IFN-γ治疗组,Smad2/3蛋白阳性表达面积均低于空白对照组(P<0.05)以及正常对照组(P<0.01),其中联合治疗组的Smad2/3蛋白阳性表达面积又明显低于吡喹酮组(P<0.05);空白对照组的Smad4蛋白的阳性表达面积高于正常对照组(P<0.01),而经吡喹酮单独治疗或吡喹酮与IFN-γ联合治疗后,Smad4蛋白阳性表达面积较空白对照组均有所下降(P<0.05)并达到正常对照组水平(P>0.01);空白对照组和吡喹酮组Smad7蛋白阳性表达面积与正常对照组比较无显著性差异(P>0.01),但在吡喹酮联合IFN-γ治疗组,Smad7蛋白的阳性表达面积明显高于其他三组(P<0.05)。结论经吡喹酮联合IFN-γ治疗后,日本血吸虫性肝纤维化小鼠肝组织中Smad2/3蛋白和Smad4蛋白表达下降,而Smad7表达上调。
