摘要
目的观察自发性高血压大鼠(SHR)肥厚的左心室心肌组织微小RNA-1(miR-1)、微小RNA-133(miR-133)表达的变化。方法选取血压升高合并左心室肥厚(LVH)的17周龄雄性SHR为LVH组(n=10),以出现血压升高无LVH的8周龄雄性SHR作为高血压组(n=10),17周龄雄性WKY大鼠作为对照组(n=10)。通过HE染色,观察大鼠左心室心肌组织病理学改变,提取大鼠左心室心肌组织总RNA,实时荧光定量聚合酶链反应(qRT-PCR)检测miR-1和miR-133在SHR肥厚左心室心肌中的表达。结果与对照组比较,LVH组和高血压组的收缩压、舒张压升高[收缩压(188.5±13.6),(152.3±10.8)比(130.6±10.1)mm Hg,P<0.01;舒张压(123.5±15.3),(94.3±10.4)比(74.3±17.4)mm Hg,P<0.05]。对照组和高血压组左心室质量(LVM)/体质量及心肌细胞横径差异无统计学意义(均P>0.05);与高血压组比较,LVH组左心室心肌细胞肥大,心肌间质增多,LVM、LVM/体质量及左心室心肌细胞横径增大[分别为(956±33)比(684±24)mg,(2.87±0.15)比(2.17±0.14)mg/g,(15.32±0.61)比(12.01±0.43)μm,均P<0.05],左心室心肌组织miR-1表达升高,miR-133表达降低。结论SHR肥厚左心室心肌中的miR-1和miR-133在不同时期出现差异表达,表现为miR-1表达升高,miR-133表达降低。
Objective To investigate the differences in the expressions of miRNA-1(miR-1)and miRNA-133(miR-133)in the tissue of hypertrophic left ventricle of spontaneously hypertensive rats(SHR). Methods17-week-old male SHR with left ventricle hypertrophy(LVH group,n=10),8-week-old male SHR without LVH(SHR group,n=10)and 17-week-old male Wistar-Kyoto(WKY)rats(WKY group,n=10)were used in this study. HE staining was used to measure the pathological changes and quantitative real-time PCR(qRT-PCR)used to determine the expressions of miR-1and miR-133 in the left ventricular tissue. Results Compared with WKY group,systolic blood pressure(SBP)and diastolic blood pressure(DBP)were higher in LVH group and SHR group[(188.5±13.6),(152.3±10.8)vs(130.6±10.1)mm Hg,P0.01;(123.5±15.3),(94.3±10.4)vs(74.3±17.4)mm Hg;P0.05]. There were no statistical differences in left ventricular mass(LVM)/body mass and the transverse diameter of myocardial cells between WKY group and SHR group(all P0.05). Compared with SHR group,left ventricular myocardial cells became marked hypertrophy,myocardial interstitium increased,LVM,LVM/body mass and transverse diameter of myocardium were larger in LVH group[(956±33)vs(684±24)mg,(2.87±0.15)vs(2.17±0.14)mg/g,(15.32±0.61)vs(12.01±0.43)μm,all P0.05]. In addition,the level of miR-1 significantly increased and the level of miR-133 significantly decreased in LVH group.Conclusion Different expressions of miR-1and miR-133 were detected in the left hypertrophic ventricular tissue of SHR and the level of miR-1significantly increased while the level of miR-133 significantly decreased.
