摘要 探讨PFOS对雄性小鼠睾丸细胞凋亡以及相关基因表达的影响。40只雄性昆明系小鼠随机分成PFOS 0、1.25、2.5、5.0和10.0 mg/kg,共5组,通过饮水方法染毒35天。处死小鼠,取睾丸,流式细胞仪观察细胞凋亡RT-PCR观察Bax和Bcl-2基因表达。细胞凋亡率随着PFOS的浓度增加而增加,各剂量组与对照组比较均有显著性差异;10mg/kg/d PFOS实验组小鼠睾丸组织中Bax基因表达与对照组比较显著上升(p<0.05),其他剂量组变化与对照组比较差异无统计学意义(p>0.05)。随着染毒剂量的增加,Bcl-2基因表达下降,5.0、10.0 mg/kg/d PFOS实验组与对照组相比差异有统计学意义(分别为p<0.05,p<0.01)PFOS能够引起睾丸细胞凋亡增加,Bax和Bcl基因表达改变可能是其机制之一。 To detect the effect of PFOS on the apoptosis and the express of Bax and Bcl-2 in testes of male mice. 40 male mice were divided into five groups, and were exposed to 0, 1.25, 2.5, 5.0 和 10.0 mg/kg PFOS. After 35 days, mice were sufficed and testes were tested by flow cytometry and the gene express of Bcl-2 and Bax were detected by RT-PCR. The rate of apoptosis increased with the increase of PFOS and there is a significant difference between the PFOS-treated groups and the control group. The gene express of Bax of 10mg/kg PFOS is significantly different from the control group (p〈0.05) and there is no difference between other group and the control group (p〉0.05).The gene express of Bcl-2 increased with the increase of PFOS. There is significant difference in group of 5 mg/kg and 10mg/kg PFOS compared to the control group. PFOS can induce the increase of apoptosis of testes and the effect of PFOS on the express of Bcl-2 and Bax gene may be the one of mechanisms.
