重组腺病毒介导的ING-4用于人MCF-7乳腺癌细胞的体外基因治疗

摘要 目的研究重组腺病毒介导的ING-4感染人MCF-7乳腺癌细胞后对其的生长抑制作用。方法将搭载有ING-4基因的重组腺病毒载体Ad-ING-4感染人MCF-7乳腺癌细胞,用荧光显微镜、RTPCR和Western-Blot法检测ING-4在MCF-7细胞中的转录和表达;CCK法与流式细胞技术检测ING-4基因对MCF-7细胞的生长抑制和促凋亡作用;半定量RT-PCR法检测ING-4基因表达对MCF-7细胞中相关凋亡基因表达的影响。结果在MCF-7细胞中ING-4基因的表达对其细胞增殖有明显抑制作用,并显著促进其凋亡,ING-4表达使MCF-7细胞中Bax表达上调,Bcl-2、Survivin表达下调。结论 MCF-7细胞在转染ING-4基因后其增殖受到了明显抑制,且更易凋亡,可能是通过改变Bax,Bcl-2及Survivin表达水平来实现。 Objective To explore the growth-inhibitory effects on the MCF-7 human breast cancer cells of the adenovirus-mediated ING-4 gene. Methods The MCF-7 human breast cancer cells were infected with AdING-4. Fluorescence microscope,RT-PCR and Western-Blot analysis were selected to detect the transcription of ING-4 gene. Cell growth inhibition and apoptosis were detected by CCK-8 assay and flow cytometry. In addition,RT-PCR was selected to detect the expression of Bax and Bcl-2. Results The cell-growth of MCF-7 cells infected by Ad-ING-4 were obviously inhibited,and the infected cells had a higher apoptosis rate. Expression of ING-4 had relationships with the up-regulation of Bax and down-regulation of Bcl-2 and Survivin.Conclusions Ad ING4 can inhibit the growth of MCF-7 cells and induce cell apoptosis,which may result in changing Bax and Bcl-2 expression.
