摘要
比较人肝癌细胞系HepG2细胞感染和未感染HEV后细胞内蛋白质表达谱的变化,为初步探索HEV在宿主细胞内感染机制及致病机理奠定基础。方法RT.nPCR和免疫荧光确认HEV感染HepG2细胞后.利用同位素标记相对和绝对定量(isobarictagsforrelativeandabsolutequantitation,iTRAQ)技术比较HEV感染组与未感染组HepG2细胞蛋白质表达差异。结果HEV感染导致328种蛋白发生变化,与未感染组相比,262种蛋白表达上调,66种蛋白表达下调。结论HEV感染HepG2细胞导致大量蛋白差异表达.功能预测表明这些蛋白涉及到宿主的物质代谢、细胞信号转导、免疫蛋白、细胞骨架成分等方面,为今后研究HEV感染机制和致病机理奠定基础。
Objective To compare the protein expression patterns of HepG2 cells infected with hepatitis E virus (HEV) or not in order to lay a foundation for studies on the mechanism and path- ogenesis of HEV infection in host ceils. Methods The differential expression of proteins was com- pared between HepG2 ceils infected with HEV or not by using isobaric tags for relative and absolute quantitation (iTRAQ) after HEV infection in HepG2 cells was conformed by reverse transcription- nested PCR (RT-nPCR) and immunofluorescenee assay (IFA) . Results A total of 328 proteins were significantly changed in HEV-infected HepG2 ceils compared with non-infected cells, inclu- ding up-regulated 262 proteins and down-regulated 66 proteins. Conclusion HEV infection resulted in the differential expressions of a large number of proteins in HepG2 cells. Function prediction showed that the proteins changed in HEV-infected HepG2 cells were involved in the metabolism and cell signaling, and were associated with immune proteins and cytoskeletal components. The current study lays a foundation for further studies on the mechanism and pathogenesis of HEV infection.
