人毛囊干细胞Notch信号通路阻滞作用研究 被引量:2

摘要 目的 研究Notch信号通路阻滞对人毛囊干细胞增殖分化的调节作用. 方法 收集2012年1月-2013年12月整形外科手术残留的头部皮肤组织,通过显微分离结合酶消化法分离人毛囊隆突区细胞,应用不同浓度的Notch信号通路阻滞剂DAPT处理后,观察对细胞的克隆形成能力、细胞周期及细胞凋亡,并进行分析. 结果 Notch信号阻断后,对人毛囊干细胞的增殖、分化及凋亡具有一定的调节作用,其中20 μmol/L药物处理组克隆形成率提高到(84±12)%,细胞总凋亡比例下降为3.81%.Western blot结果示N1ICD、Hes1及p21表达均有降低,Wnt-10b的表达上调. 结论 Notch信号通路阻滞剂DAPT能有效提高人毛囊干细胞比例,减少细胞凋亡,其调节机制可能与其对p21表达的调控以及与Wnt通路的交互作用有关.
