摘要
目的观察热休克蛋白27(HSP27)在砷致Ha Ca T恶性转化细胞中的水平变化情况,探讨其在砷致Ha Ca T细胞恶性转变中的作用。方法用三氧化二砷作为诱导剂,建立Ha Ca T细胞恶性转化模型,用RT-PCR方法检测HSP27mRNA的表达情况,用Western blot法检测HSP27蛋白水平。结果 Ha Ca T恶性转化细胞HSP27mRNA表达和蛋白水平均较对照组上调,差异均有统计学意义(P﹤0.05)。结论Ha Ca T细胞长期在含三氧化二砷培养基中培养可使HSP27mRNA表达和蛋白水平均上调,提示HSP27在砷诱导Ha Ca T细胞恶性转变过程中发挥着一定的作用。
Objective To investgate the expression levels of HSP27 during arsenic-induced malignant transformation of human HaCaT ceils and its significance. Methods The HaCaT cells were exposed chronically to a low level of arsenic to get malignant phenotype. The level of HSP27 mRNA was assessed by RT-PCR method. The protein level of HSP27 was determined by Western blotting method. Results The HSP27 mRNA expression level and the protein level of HSP27 in HaCaT malignant transformation cells were significantly higher than that in the HaCaT cells of control group, the difference is statistically significant ( P 〈 0.05 ). Conclusion HaCaT ceils long-term cultured in mediums contain arsenic may show up-regulation of HSP27 mRNA and HSP27 protein of the cells, suggesting that HSP27 may play a role in the malignant transformation of HaCaT cells induced by arsenic.
