矢车菊素-3-葡萄糖苷对高胆固醇血症小鼠血小板颗粒物释放的影响 被引量:12

The effects of cyanidin-3-glucoside on platelet granule secretion in hypercholesterolemia mice

摘要 目的探讨植物花色苷矢车菊素-3-葡萄糖苷(Cy-3-g)对高胆固醇血症小鼠血小板颗粒物释放的影响。方法将7周龄雄性C57小鼠随机分成正常对照组、高胆固醇血症模型组和Cy-3-g干预组。正常对照组饲喂AIN-93G标准饲料,高胆固醇模型组饲喂高脂高胆固醇饲料,Cy-3-g干预组饲喂添加Cy-3-g(1 000 mg/kg)的高脂高胆固醇饲料。饲喂12周后,取小鼠血液测定血清总胆固醇(TC)的含量。确认模型建立成功后,心脏取血,分离纯化血小板悬液,测定各组小鼠血小板的三磷酸腺苷(ATP)释放、五羟色胺(5-HT)释放以及P选择素(CD62P)和CD40L在血小板表面的表达量。结果实验期末,高胆固醇血症组和Cy-3-g干预组的血清总胆固醇明显高于正常对照组。与高胆固醇血症组相比,干预组血小板的ATP释放、5-HT释放及CD62P和CD40L表达水平明显降低。结论花色苷Cy-3-g能够抑制高胆固醇血症小鼠血小板颗粒物释放,提示植物花色苷对心血管疾病具有一定的保护作用。 Objective To investigate the effects of plant anthocyanin cyanidin-3-glucoside(Cy-3-g) on platelet granule secretion in hypercholesterolemia mice. Methods The mice were divided into control, hypercholesterolemia and treatment groups, feeding with AIN-93 G, hypercholesterolmic diets, and hypercholesterolemic diets added with Cy-3-g(1 000 mg / kg),respectively. After 12 weeks, blood samples were taken by cardiac aspiration after the mice were anesthetized. To assure differently elevated cholesterol levels, the level of total cholesterol(TC) in serum was determined. Then ATP, 5-HT, CD62 P and CD40 L releasing were examined as platelet granule secretion. Results The TC levels on hypercholesterolemia and treatment groups were significantly higher than that of control group. After stimulated with thrombin, levels of ATP and 5-HT releasing from platelets, CD62 P and CD40 L expressed on platelets were significantly less than those of hypercholesterolemia group. Conclusion Anthocyanin Cy-3-g can inhibit platelet granule secretion, indicating health care function of anthocyanin on cardiovascular diseases.
