大承气汤对重症急性胰腺炎大鼠肠道免疫屏障的保护作用 被引量:13

Protective Effect of Dachengqi Decoction on Intestinal Immune Barrier of Rat with Severe Acute Pancreatitis

摘要 目的探讨大承气汤对重症急性胰腺炎(SAP)大鼠肠道免疫屏障的保护作用。方法大鼠随机分为假手术组、SAP组和大承气汤组。假手术组不给予药物干预,SAP组和大承气汤组于造模前24h、18h、12h、6h分别给予蒸馏水和大承气汤灌胃。SAP大鼠模型采用逆行胆胰管注射法制备。各组动物于造模后6h、12h、24h、36h取腹主动脉血和末端回肠组织,血清生化法测定淀粉酶,RT-PCR法检测末端回肠组织的HMGB1-RNA和COX-2 RNA表达。结果 SAP组和大承气汤组各时间点死亡比较,大承气汤组的死亡率在24h、36h较低。在造模后6h,3组动物的淀粉酶、HMGB1-RNA表达、COX-2 RNA表达均达到最高,但假手术组最低,其次为大承气汤组;在12h、18h、24h时,与SAP组比较,大承气汤组的淀粉酶、HMGB1-RNA表达、COX-2 RNA表达能够较快地降低。结论大承气汤能够降低SAP大鼠末端回肠的HMGB1-RNA和COX-2 RNA表达,从而保护肠道免疫屏障。 Objective To observe protective effect of Dachengqi decoction on intestinal immune barrier of rat with severe acute pancreatitis( SAP). Methods 120 rats were randomly divided into sham group,SAP group and Dachengqitang group. Sham group were not given drug. SAP group and Dachengqitang group were respectively gavage by distilled water and Dachengqitang on 24 h,18 h,12 h and 6 h before surgery. SAP model was prepared by retrograde cholangiopancreatography. Experimental animals were taken abdominal aorta blood and distal ileum on 6 h,12 h,24 h,and 36 h after surgery. Amylase in serum and the expression of HMGB1- RNA and COX- 2 RNA in distal ileum were studied. Results Dachengqitang group had lower mortality than SAP group on 24 h,36h. Serum amylase levels and the levels of HMGB1- RNA and COX- 2 RNA expression were peaked. Among the three groups,sham group was the lowest,followed by the Dachengqitang group. Dachengqitang treatment significantly decreased serum amylase levels and the levels of HMGB1- RNA and COX- 2 RNA expression. Conclusions Dachengqi decoction could protect the intestinal immune barrier,with ameliorating the levels of HMGB1- RNA and COX- 2 RNA expression.
