摘要
不同的大豆品种对于农杆菌的敏感性不同,易感型的品种更有利于转化的进行。为了进一步优化大豆转化体系,实验利用农杆菌菌株EHA105、GV3101对东农50等10个大豆栽培品种进行敏感性测定。同时对2个易感性品种,东农50、中豆32共培养中添加光照处理,统计GUS染色率。结果表明,农杆菌GV3101的侵染能力较强,10个品种的平均GUS染色率达到66.7%,远远高于EHA105的平均值(38.4%);2个菌株对中豆32和东农50进行侵染,GUS染色率平均达到69.93%和73.62%,其他品种的GUS平均染色率显著低于东农50和中豆32;但个别品种对2个菌株的敏感性差异显著,在大豆遗传转化中对于品种和菌株的选择具有重要的意义。农杆菌的侵染时间为30~60min时效果较好;在共培养阶段进行光照处理有利于提高转化率。
Ten varieties and 2 Agrobacterium GV3101 and EHA105 were used to study factors,such as genotypes of soybean,strains and concentrations of Agrobacteriumand adding light condition in co-cultural period,affecting the soybean transformation.The results revealed that the infection efficiency of Agrobacterium GV3101 is 68%,which is higher than that of Agrobacterium EHA105(38.4%).Dongnong50 and Zhongdou 32 are more sensitive as the receptors to Agrobacterium GV3101 and EHA105in transformation.When explants were inoculated with Agrobacteriumfor 30 to 60mins,transient expression efficiency of GUS is higher.Putting explants in the culture condition of 16 hours lighting instead of all in dark in the cocultivation period,GUS transient expression is increased.
