黄芪白花蛇舌草汤对阿霉素肾病大鼠的治疗作用

Therapeutic effects of Astragalus-diffusa decoction on adriamycin-induced nephropathy rats

目的观察黄芪白花蛇舌草汤对肾病综合征(NS)大鼠的治疗作用。方法健康雄性SD大鼠40只,随机抽取10只作为正常对照组,其余大鼠一次性尾静脉注射盐酸阿霉素6 mg/kg,建立肾病综合征大鼠模型。造模成功后随机分为3组:模型组,黄芪白花蛇舌草汤低剂量组3.6g/(kg·d),黄芪白花蛇舌草汤高剂量组70.2 g/(kg·d)。连续灌胃给药4周,观察黄芪白花蛇舌草汤对肾病综合征大鼠的24 h尿蛋白、血生化指标、肾组织病理学等方面的影响。结果与模型组比较,各治疗组大鼠24 h尿蛋白定量明显降低(P<0.05),总蛋白、白蛋白升高(P<0.05),总胆固醇、三酰甘油降低(P<0.05)。结论黄芪白花蛇舌草汤具有降低尿蛋白、降血脂、提高血清蛋白等作用,改善病理组织学变化,减轻肾脏损害,对肾病综合征大鼠具有良好的治疗作用。

Objective To investigate the therapeutic effects of Astragalus-diffusa decoction on Adriamycin-induced nephropathy（ NS） rats. Methods The 40 healthy male SD rats,in which 10 were randomly selected as the normal control group. Nephritic syndrome models were established in 30 rats by intravenous injection of Adriamycin 6. 0 mg / kg.After the modeling,they were randomly divided into three groups： model control group,low-dosage group 3. 6 g /（ kg·d） and high-dosage group 70. 2 g /（ kg·d）. The rats of low-dosage group and high-dosage group were intragastric administrated decoction of Astragalus-diffusa soup daily,the model control group rats were given running water. During 4weeks treatment process,the 24 hour urine protein concentration of rats were observed. The level of total serum protein（ TP）,serum albumin（ ALP）,total cholesterol（ TCH）,triglycerides（ TG） and UREA were determined,and the pathological changes of kidney tissue were assayed after the therapy process. Results After 4 weeks therapy,the 24 hour urine protein concentration of low-dosage group and high-dosage group were significantly lower than model control（ P〈0. 05）. The level of TP and ALP of low-dosage group were significantly higher than model control（ P〈0. 05）,as well as the level of TCH and TG were significantly lower（ P〈0. 05）. Conclusion Astragalus-diffusa decoction could decrease the urine protein concentration and improves renal pathology to protect the kidney from damage by reducing the serum cholesterol level,decreased blood lipid content,and increased the plasma protein level.
