A群轮状病毒NSP5蛋白在细胞器中定位特性 被引量:1

摘要 为研究A组猪轮状病毒(Po RV)非结构蛋白NSP5在MA-104细胞中的定位情况,本研究将NSP5蛋白基因克隆于表达载体p EGFP-C2中构建重组质粒p EGFP-NSP5,将其转染于MA-104细胞中进行NSP5瞬时表达,并于转染48 h后利用激光共聚焦显微镜分析NSP5表达蛋白在细胞中的定位情况。结果表明,表达蛋白在MA-104细胞中的线粒体、内质网、高尔基体中均有分布,主要定位于细胞质。本研究为进一步分析病毒的复制和致病机理提供依据。 To study the localization of A group porcine rotavirus (PoRV) non-structural protein NSP5 in the cells, MA-104 cells were transfected with recombinant plasmids of pEGFP-NSP5 expressing NSP5 protein and examined using eonfocal microscopy at 48 hours post transfection. The results showed that the NSP5 protein localized both in mitochondrion, endoplasmic reticulum and golgi apparatus in the transfected cells, suggesting NSP5 protein had the ability of localization in cytoplasm, which provided the basis for further studies of PoRV replication and pathogenesis mechanism.
