摘要 目的:检测C/EBPα在结肠直肠癌中的表达情况,探讨下调C/EBPα表达对结肠癌细胞增殖、凋亡以及细胞周期的影响。方法:利用RT-PCR和免疫组织化学技术检测C/EBPα在结肠直肠癌组织及相邻正常组织中的表达,分析C/EBPα表达与临床病理特征的关系。用Western印迹法筛选C/EBPα,发现SW620为高表达C/EBPα肠癌细胞株。通过RNA干扰技术下调结肠癌细胞株SW620中C/EBPα表达后,利用CCK8和流式分析技术检测SW620细胞增殖、凋亡和细胞周期的变化。结果:C/EBPα在结肠直肠癌组织中高表达,且与肿瘤浸润深度有关(P<0.05);下调C/EBPα表达抑制SW620细胞株增殖(P<0.05),促进SW620细胞凋亡(P<0.05),并使SW620细胞阻滞于细胞周期的S期。结论:C/EBPα在结肠直肠癌组织中高表达,下调C/EBPα可抑制结肠癌细胞增殖,促进其凋亡,阻滞细胞周期。 Objective To investigate the expression of CCATT/enhancer binding protein alpha(C/EBPα) in colorectal cancer(CRC) patients and the influence of down-regulation of C/EBPα on the proliferation, apoptosis and cell cycle of colon cancer cells. Methods The expression of C/EBPα in both tumor tissues and adjacent normal tissues of the patients with CRC were assessed by RT-PCR and immunohistochemistry. The association of expression of C/EBPα with clinicopathological parameters was analyzed. Colon cancer cell lines were screened for expression of C/EBP α by Western blot. SW620 colon cancer cells were found higher expressed C/EBPα and transfected with C/EBPα-sh RNAs. The effects of down-regulated C/EBPα on cell proliferation, apoptosis and cell cycle were analyzed by CCK-8 and flow cytometry, respectively. Results It was showed that C/EBPα was highly expressed in CRC tissues and was associated with tumor invasive depth(P〈0.05). Silencing C/EBPα in SW620 colon cancer cells suppressed cell proliferation, increased cell apoptosis and induced cell cycle arrest(P〈0.05). Conclusions C/EBPα was highly expressed in human CRC tissues. Down-regulation of C/EBPα suppressed cell proliferation, increased cell apoptosis and induced cell cycle arrest.
