摘要
目的探讨nm23-H1基因转染对肺癌细胞株基质金属蛋白酶9(MMP-9)和金属蛋白酶组织抑制物1(TIMP-1)表达的影响。方法利用脂质体介导的基因转染技术,将nm23-H1基因转染入肺癌细胞株L9981;利用半定量RT-PCR技术检测nm23-H1基因转染前后L9981肺癌细胞株TIMP-1、MMP-9基因的mRNA表达;利用流式细胞仪技术检测nm23-H1基因转染前后L9981肺癌细胞株TIMP-1、MMP-9基因的蛋白表达。结果 nm23-H1基因转染后肺癌细胞株TIMP-1 mRNA及蛋白表达上调,MMP-9 mRNA及蛋白表达下调。结论 nm23-H1基因对L9981肺癌细胞株TIMP-1 mRNA与蛋白表达具有正向调控作用,对MMP-9 mRNA与蛋白表达具有负向调控作用。
Objective To investigate the influence of nm23-H1 gene transfection on the expressions of matrix metalloproterinase9(MMP-9)and tissue inhibitor of metalloproteinase 1(TIMP-1) in lung cancer cells.Methods nm23-H1 gene was transfected into lungcancer cell line L9981 by gene transfection technology. Semi-RT-PCR was used to detect the different mRNA expressions of TIMP-1 and MMP-9 between transfected and non-transfected lung cancer cell lines. Flow cytometry was used to detect the different protein expressionsof TIMP-1 and MMP-9 between transfected and non-transfected lung cancer cell lines.Results The mRNA and protein levels of TIMP-1 in transfected lung cancer cell lines were stronger than those in non-transfected lung cancer cell lines. The mRNA and protein levels of MMP-9 in transfected lung cancer cell lines were weaker than those in non-transfected lung cancer cell lines.Conclusion The nm23-H1 gene can up-regulate expressions of TIMP-1 and down-regulate expressions of MMP-9 in lung cancer cell line L9981.
