摘要
目的:观察脊髓损伤后大鼠逼尿肌反射亢进膀胱Cajal间质细胞(ICC)细胞兴奋性的变化,探讨膀胱逼尿肌功能改变与ICC细胞兴奋性变化的可能关系。方法:制作大鼠骶髓上损伤模型,膀胱压力测定证实膀胱出现逼尿肌反射亢进后,取膀胱组织急性分离ICC细胞,激光共聚焦显微镜观察ICC细胞兴奋性变化。结果:逼尿肌反射亢进大鼠模型制作成功,膀胱ICC细胞兴奋性较正常组和膀胱造瘘组明显增加。结论:脊髓损伤后大鼠膀胱功能改变可能与ICC细胞兴奋性增加有关。
Objective:To investigate the excitability change of interstitial cells of Cajal(ICC)in rat bladder with detrusor hyperreflexia after spinal cord injury.Method:The rat model with suprasacral spinal cord injury(SSCI)was made,and cystometry were conducted to confirm that bladder showed detrusor hyperreflexia after spinal cord injury.The ICCs of bladder with detrusor hyperreflexia were freshly dispersed and the spontaneous calcium waves of ICCs were detected using laser confocal microscopy.Result:The model of rat with detrusor hyperreflexia was made successfully.The spontaneous excitability of ICCs in SSCI rats was higher than that in normal and post-cystostomy rats(P〈0.05).Conclusion:The functional change of bladder may be associated with the excitability change of ICC in rat after spinal cord injury.
